Plantlets were regenerated from leaf explants of Actinidia eriantha Benth. cv White (kiwifruit). The best medium for adventitious shoot formation from leaf blade strips was Murashige-Skoog (MS) medium with 0.1 mg L(1 NAA and 2.0 mg L (1 BA. The frequency of shoot regeneration (%) was dependent on the size and the part of the leaf from which the explant was taken. The highest frequency of shoot regeneration was 7.3092.61 shoots per explant, for explants taken from within 0Á3 cm of the leaf blade base, near a vein and not on the leaf edge. When using petiole-derived explants, a maximum frequency of adventitious shoot regeneration of 79.17% with 6.5492.31 shoots per explant was obtained using 0.1 mg L(1 NAA and 2.0 mg L (1 ZT in MS medium. The highest rate of rooting obtained was 96.67% when using 1/2 MS medium containing 0.6 mg L(1 IBA and 93.75% when using wet moss without IBA. This is the first report to study the effect of explants taken from different parts of the leaf blade on shoot regeneration in A. eriantha. All plantlets were transferred to pots of peat/perlite and grown on before being planted in the field. Plantlets were successfully acclimatized to field conditions and produced healthy plants.

Changes in the tubulin cytoskeleton during protoplast culture and plant regeneration of Solanum lycopersicoides Dun. were analyzed using an immunodetection method. Directly after isolation, four groups of protoplasts were distinguished: (1) mononuclear, (2) polynuclear, (3) homogeneous, (4) anuclear. The tubulin cytoskeleton of the protoplasts underwent rearrangements, correlating to the number and structure of cell nuclei in the protoplast. All protoplast groups with the exception of mononuclear were characterized by perturbations in the organization of the tubulin cytoskeleton. Anuclear and homogeneous protoplasts did not have a tubulin cytoskeleton. Polynuclear protoplasts had cortical microtubules, but were not capable of re-forming their original arrangement and did not possess a radial or perinuclear cytoskeleton. Irregularities in microtubule arrangement of these three groups of protoplasts caused their inability to regenerate a cell wall and to divide. Anuclear, polynuclear and homogeneous protoplasts were eliminated from the culture. Mononuclear protoplasts rearranged their cortical microtubules and reestablished the radial and perinuclear tubulin cytoskeleton. Re-formation of the cell suspension and subsequent regeneration of plants occurred exclusively from mononuclear protoplasts, which were able to regenerate cell walls and to divide.

Rapid Shoot Propagation from Micro-Cross Sections of Kiwifruit (Actinidia deliciosa cv. ‘Hayward’)

Kiwifruit shoots can be rapidly propagated through a micro-cross section (MCS) system we established here. Optimal culture conditions were determined for different explant types, section sizes, and concentrations of inorganic salts and plant growth regulators. Rates of survival and callus formation were higher in half-strength MS salts than in full-strength MS media. Similar performance (i.e., survival and callus formation) was achieved with section sizes of either 800 pm or 1200 pm. Proliferation efficiency was greatest when explants from stem tissue were cultured on 1/2 MS supplemented with 4.5x10 3 pM 2,4-dichlorophenoxyacetic acid and 4.6x10 i pM zeatin. The number of shoots averaged 2.61 per explant, representing an efficiency of 94%. RAPD analysis revealed that the regenerated plants from our MCS system were genetically stable. These results show that the culturing of micro-cross sections from stem tissue is a powerful method for kiwifruit propagation.
