Pharmacological basis for sedative and hypnotic like effects of Pyrus pashia using in vivo experimental models

Objective Pyrus pashia has traditionally been used as a therapeutic agent including sedative. In this regard, hydroethanolic extract of Pyrus pashia (HEPP) was screened for phytochemical investigation, acute toxicity, and sedative‐hypnotic activity to provide a scientific rationale to its ethno‐medicinal uses. Methods Mice were used in acute toxicity test; sedative potential was observed in open field test while thiopental‐induced sleeping time and barbiturate‐induced sleeping time tests were used for the assessment of hypnotic like effects of HEPP. Various phytochemical groups were detected in preliminary phytochemical tests. Results In acute toxicity study, the extract was found safe up to 1g/kg while the extract caused 50% death at 1500 mg/kg. In all animal models, the extract had shown sedative and hypnotic effects but the most significant effect observed at increased dose in comparison with standard diazepam‐treated group. In open field test, the extract at doses of 200 and 400 mg/kg, there is mild decrease in number of lines crossed by tested animals (mice) while significant reduction shown at 600 mg/kg compared with diazepam‐treated group at dose 0.5 mg/kg. In phytochemical investigation HEPP subjected to different phytochemical tests. From the thoroughly study, it is revealed that these constituents mainly participating in sedation and hypnosis. Conclusions The results of the present study showed significant sedative‐hypnotic effects of the HEPP in mice and that the central benzodiazepine receptors are involved in the sedative‐hypnotic effects of this plant.

Effects of alkaloids on peripheral neuropathic pain: a review

Neuropathic pain is a debilitating pathological pain condition with a great therapeutic challenge in clinical practice. Currently used analgesics produce deleterious side effects. Therefore, it is necessary to investigate alternative medicines for neuropathic pain. Chinese herbal medicines have been widely used in treating intractable pain. Compelling evidence revealed that the bioactive alkaloids of Chinese herbal medicines stand out in developing novel drugs for neuropathic pain due to multiple targets and satisfactory efficacy. In this review, we summarize the recent progress in the research of analgesic effects of 20 alkaloids components for peripheral neuropathic pain and highlight the potential underlying molecular mechanisms. We also point out the opportunities and challenges of the current studies and shed light on further in-depth pharmacological and toxicological studies of these bioactive alkaloids. In conclusion, the alkaloids hold broad prospects and have the potentials to be novel drugs for treating neuropathic pain. This review provides a theoretical basis for further applying some alkaloids in clinical trials and developing new drugs of neuropathic pain.

Daun pepaya memiliki kandungan beberapa senyawa alkaloid (karpain, pseudokarpain, dehydrokarpain I, dehydrokarpain II, dan emetin). Penelitian ini bertujuan untuk mengetahui aktivitas antiplatelet, antikoagulan, dan trombolitik alkaloid total daun pepaya secara in vitro. Penelitian ini terdiri dari 5 perlakuan: kontrol negatif, kontrol positif, alkaloid total daun pepaya (konsentrasi 0.5, 1.0, dan 2.0 mg/mL) dengan parameter persentasi inhibisi agregasi, persentase inhibisi koagulasi dari nilai Clotting Time (CT), Prothrombine Time (PT), dan Activated Parsial Thromboplastine Time (APTT), dan daya fibrinolitik. Berdasarkan analisis statistik diketahui bahwa terdapat perbedaan yang signifikan antara persentase inhibisi agregasi alkaloid total dibanding kontrol negative, namun tidak berbeda dengan kontrol positif (clopidogrel). Pada uji antikoagulan, alkaloid total daun pepaya secara signifikan dapat memperpanjang CT, PT, dan APTT yang berbeda signifikan dengan kontrol negatif, namun tidak berbeda dengan kontrol positif (heparin). Hasil uji trombolitik menunjukkan bahwa alkaloid total daun pepaya dapat meningkatkan persentase trombolitik yang berbeda dengan kontrol negatif, namun tidak berbeda signifikan dengan kontrol positif (nattokinase).
