Placental Exosomes as Biomarkers for Maternal Diseases: Current Advances in Isolation, Characterization, and Detection

Abstract: Serving as the interface between fetal and maternal circulation, the placenta plays a critical role in fetal growth and development. Placental exosomes are small membrane-bound extracellular vesicles released by the placenta during pregnancy. They contain a variety of biomolecules, including lipids, proteins, and nucleic acids, which can potentially be biomarkers of maternal diseases. An increasing number of studies have demonstrated the utility of placental exosomes for the diagnosis and monitoring of patholo… Show more

“…For instance, polycarbonate membrane extrusion can collect and extrude the cells to obtain a homogeneous material through a small extruder containing a polycarbonate film. The ultrasonic crushing method usually uses the vibration force of ultrasonic waves to break up cell membranes and organelles. In this regard, the routine isolation of exosomes parallels the cell membrane extraction technique mentioned above, except that the former does not require aggressive methods such as extrusion or sonication . Overall, the maturity of these cell membrane extraction technologies has laid a solid foundation for the application of cell membranes.…”

“…Human Wharton’s jelly derived mesenchymal stem cells (hWJ-MSCs) can produce exosomes isolated with high purity via the sucrose cushion method when compared to ultracentrifugation (UC) and commercially available reagent (CR) . However, nanotechnology-based exosomes isolation is also an effective method. …”

“…Organisms produce exosomes under normal physiological and pathological conditions, and they have been detected in various bodily fluids . Exosomes mediate intercellular communication, and their contents affect various life processes, including angiogenesis, metastasis, anticancer resistance, immune evasion, macrophage polarization, and metabolic reprogramming. Exosomes are increasingly recognized as promising biomarkers for a range of diseases. Exosomes display low toxicity, biodegradability, and efficient tissue targeting, which make them reliable delivery vehicles. Although exosome therapies have been developed by some pharmaceutical companies, there are still many obstacles in the production of clinical grade exosomes for treatment . Especially, the isolation of exosomes is a crucial step in the production process of exosomes .…”
