Piwil 2 Expression Is Correlated with Disease-Specific and Progression-Free Survival of Chemotherapy-Treated Bladder Cancer Patients

Abstract: Piwi-like 2 (Piwil 2) belongs to the family of Argonaute genes/proteins. The expression of Piwil 2 is associated with stem cells. A role in tumorigenesis and/or tumor progression is proposed for different cancers but not yet for bladder cancer (BCa). We investigated the Piwil 2 expression by immunohistochemistry in a cohort of 202 BCa patients treated by cystectomy and adjuvant chemotherapy. The association between Piwil 2 expression and disease-specific (DSS) or progression-free survival (PFS) was calculated … Show more

“…The expression of PIWIL proteins varies in different tumors and, as reported recently, PIWIL1, PIWIL2, and PIWIL4 are expressed at low levels in human renal cell carcinoma and are related to shortened survival 33 . PIWIL2 has weak cytoplasmic but no nuclear expression and is associated with the progression of bladder cancer, suggesting it acts as a tumor suppressor 34 . However, PIWIL2 is up-regulated in colorectal tumors and is positively correlated with poor prognosis, indicating it acts as an oncogene 35 .…”

“…In our previous study of chemotherapy-treated bladder cancer, patients were investigated with a different Piwi-like 2 antibody, a weak cytoplasmic staining pattern (IRS 1–2) was associated with poor DSS and tumor progression 28 . The group of patients with negative Piwi-like 2 staining (IRS = 0) showed in this and in the previous study a rather good prognosis.…”

“…They found that Piwi-like 2 is not expressed in either human normal urothelial cells or bladder cancer cell lines and tissues. Previously, we showed that Piwi-like 2 expression was correlated with disease-specific and progression-free survival of chemotherapy-treated bladder cancer patients 28 . In this study, we analyzed the tumors of 95 MIBC patients for their protein expression of Piwi-like 1 and Piwi-like 2 and associated their expression with clinico-pathological and survival data.…”

“…In agreement with our observations, Taubert et al described decreased levels of PIWIL2 protein to be significantly associated with poor prognosis in cohort of 202 bladder cancer patients ( P =0.005). 20 A higher level of PIWIL4 mRNA was found to be associated with longer overall survival also in non-small cell lung cancer patients. 19 In patients with soft tissue sarcoma, Greither et al identified significant association between the low-expression levels of PIWIL2, PIWIL3, and PIWIL4 mRNAs and shorter tumor-specific survival.…”
