Abstract:Having a theoretical understanding of the orientation of immunoglobulin on an immobilized solid surface is important in biomedical pathogen-detecting systems and cellular analysis. Despite the stable adsorption of immunoglobulin on a polystyrene (PS) surface that has been applied in many kinds of immunoassays, there are many uncertainties in antibodybased clinical and biological experimental methods. To understand the binding mechanism and physicochemical interactions between immunoglobulin and the PS surface at the atomic level, we investigated the binding behavior and interactions of the monoclonal immunoglobulin G (IgG) on the PS surface using the computational method. In our docking simulation with the different arrangement of translational and rotational orientation of IgG onto the PS surface, three typical orientation patterns of the immunoglobulin G on the PS surface were found. We precisely analyzed these orientation patterns and clarified how the immunoglobulin G interacts with the PS surface at atomic scale in the beginning of the adsorption process. Major driving forces for the adsorption of IgG onto the PS surface come from serine (Ser), aspartic acid (Asp), and glutamic acid (Glu) residues.

The theorical aspects of QCM, EIS and SPR are introduced and the main application areas are outlined. INTRODUÇÃONas últimas décadas, as ciências analíticas têm experimentado um grande avanço relacionado com a capacidade de obtenção de informações químicas de objetos e sistemas. Como resultado, essas informações têm proporcionado um desenvolvimento analítico bem como sua incorporação a outras áreas científicas e técnicas, de forma a resultar na automação, miniaturização e simplificação dos sistemas 1 . Assim sendo, verifica-se o desenvolvimento e o uso apropriado de sistemas de referência, revitalização da análise qualitativa, expansão das fronteiras analíticas clássicas (amostragem, estudos in situ, interdisciplinaridade) e o desenvolvimento de sistemas analíti-cos como sensores químicos, biossensores, dispositivos bioanalíticos e eletrodos quimicamente modificados. Neste contexto, os sensores químicos e biossensores têm adquirido grande importância devido à possibilidade de aplicação destes sistemas para avaliação de processos sintéticos ou biológicos, bem como o entendimento destes processos 2 . Sensores químicos são dispositivos que transformam uma informação química, como a variação da concentração de um componente específico de uma amostra em relação à composição total, em um sinal analítico útil. Estes sistemas contêm duas unidades funcionais básicas: um receptor e um transdutor 3 . As aplicações destes no monitoramento e controle ambiental, em agricultura, indústria de alimentos, farmacêutica e clínica têm possibilitado análises com monitoramento contínuo e in vivo em tempo real 4 . O desenvolvimento de dispositivos sensores tem como premissa básica a avaliação de processos interfaciais e superficiais, uma vez que estes processos são de fundamental importância no entendimento dos mecanismos de interação entre o sistema de reconhecimento e o analito 5,6 . Além disso, o estudo de processos interfaciais e superficiais é de grande relevância para a escolha do método de imobilização de espécies sobre a superfície de eletrodos e a avaliação de problemas associados aos métodos de imobilização. Ahstraet: The detection of anti-human immunodefficiency virus (HIV) antibodies by means of synthetic HIV peptide immobilized on a piezoelectric quartz sensor is demonstrated The measurement set-up consists of an oscillator circuit, a suitably modified AT-cut thickness-shear-mode quartz crystal with gold electrodes, which is housed in a special reaction vessel, and a computercontrolled frequency counter for the registration of the measured frequency values. The quartz crystal is adapted for a steady operation in liquids at a frequency of 20 MHz. In phosphate-buffered saline solution the oscillator reaches a stability of about 0.5 Hz within a few seconds, of about 2 Hz within 10 min and about 30 Hz within 1 h. The frequency shift due to the adsorption of various proteins to the uncoated sensor surface has been investigated. It can be shown that a stable adsorptive binding of proteins to an oscillating gold surface is feasible and can be used for the immobilization of a receptor layer (e.g. HIV peptide). Specific binding of the anti-HIV monoclonal antibody to the HIV peptide immobilized on the quartz sensor is demonstrated. Control experiments show, however, additional unspecific binding. According to the experiments, the Sauerbrey formula gives a sufficiently accurate value for the decrease of the resonant frequency due to adsorption or binding of macromolecular proteins on the quartz crystal surface.
