PI3K/AKT/mTOR signaling transduction pathway and targeted therapies in cancer

Antonino Glaviano,
Aaron S. C. Foo,
Hiu Y. Lam
et al.

Abstract: The PI3K/AKT/mTOR (PAM) signaling pathway is a highly conserved signal transduction network in eukaryotic cells that promotes cell survival, cell growth, and cell cycle progression. Growth factor signalling to transcription factors in the PAM axis is highly regulated by multiple cross-interactions with several other signaling pathways, and dysregulation of signal transduction can predispose to cancer development. The PAM axis is the most frequently activated signaling pathway in human cancer and is often impli… Show more

“…Although we did not find extra downstream signaling pathways that were highly enriched, the changes in transduction may still have an effect on the downstream mTOR signaling pathway (Figure ), which is essential for glycerolipid metabolism, autophagy control, and protein synthesis . Collectively, our results support the idea that changes in this group of signaling mechanisms may contribute to CW formation.…”

“…In addition, the downstream effectors of AKT/mTOR and EGFR signalling are also correlated to CSC-associated survival and metastasis in several cancers (Glaviano et al, 2023). Studies have also shown that Silibinin-treated colon CSCs decreased the fraction of CD44+ surface markers and prevented the development of spheres by boosting the inactivation of the AKTSer473/mTOR survival pathways, suggesting its role in negatively regulating tumour growth (Wang et al, 2012).…”

“…Notably, vascular endothelial cells demonstrate a remarkable capability for rapid proliferation, migration, and differentiation in response to physiological stimuli 25 . VEGF has the ability to activate numerous downstream pathways, such as the PI3K-Akt/mTORC2 route, the PLC-MEK pathway, and the Src-FAK pathway that are responsible for vascular permeability, cellular proliferation and cellular motility 2628 . Through qPCR we have checked the expression of these VEGF pathway genes upon stabilization of VEGF quadruplex by de novo designed peptides.…”
