Phytochemicals: Potential Therapeutic Modulators of Radiation Induced Signaling Pathways

Abstract: Ionizing radiation results in extensive damage to biological systems. The massive amount of ionizing radiation from nuclear accidents, radiation therapy (RT), space exploration, and the nuclear battlefield leads to damage to biological systems. Radiation injuries, such as inflammation, fibrosis, and atrophy, are characterized by genomic instability, apoptosis, necrosis, and oncogenic transformation, mediated by the activation or inhibition of specific signaling pathways. Exposure of tumors or normal cells to d… Show more

“…Xanthine oxidase and lipoxygenase are inhibited by many polyphenols, thus reducing the generation of free radicals. Finally, many polyphenols decrease the activation of NF-κB and MAPK, thus reducing the release of inflammatory cytokines which play a role in the radiation-induced inflammatory response [ 102 , 103 , 104 ].…”

“…In the case of low dose radiation exposures, cancer, cardiovascular disease, foetal abnormality, and chronic fatigue are examples of gross adverse effects [ 89 ]. These are thought to result from genomic instability, mutation in critical genes, microenvironment alterations leading to niche compromise, haematological insufficiency and immune dysfunction [ 7 , 29 , 90 , 91 , 92 , 93 , 94 , 95 , 96 ]. These in turn are attributed to DNA damage, elevated ROS, mitochondrial insufficiency, membrane channel imbalances and failure of “checks and balances” on the rate of specific enzyme reactions, leading to tipping points where metabolic malfunction occurs.…”
