Background: Medicinal plants have long been utilized in traditional medicine in Turkey and worldwide. The flora of Turkey is so rich it needs to be discovered, documented, and explored. This plant species is the origin of many valuable medicinal chemicals. Salvia species was used in folk medicine for many years in Turkey. One of the new Salvia species is Salvia siirtica. Methods: The antimicrobial, cytotoxic, antioxidant, DNA restriction inhibition, and DNA protective activities of methanolic extract of S. siirtica were studied. Results: The extract has better activity than the standard antibiotics against P. aeruginosa, meanwhile, showed high DPPH scavenging activity with an IC50 value of 28.72 ± 8.02 µg/ml. The extract exhibited good cytotoxic potential with IC50 value 0.31 ± 0.07 mg/ml towards H1299 cell line. The restriction enzyme inhibition in presence of plant extract proves that extract can interact with restriction endonuclease enzymes. Apigenin and Luteolin 7- glycoside are two high components of S. siirtica were docked against potential intracellular targets to evaluate their inhibition capability. Luteolin its derivative luteolin-7-glycoside showed the highest potential in molecular docking studies. Conclusions: Determining the bioactivity of these components by isolating them from the plant may pave the way for their use as supportive agents in the fight against many diseases.

Background Nowadays, the use of herbal extracts for the production of nanoparticles has attracted a lot of attention due to the fast reaction, economy, and compatibility with the environment. The aim of the present study is the biosynthesis of silver nanoparticles from the extracts of Nepeta sessilifolia Bunge and Salvia hydrangea DC. ex Benth. and their antibacterial activity was measured. Methods For this purpose, the flowering branch of N. sessilifolia and the flower of S. hydrangea were randomly collected from three places, respectively, from the rangelands of Aqdash Mountain and Biabe in Isfahan province, Iran in May 2021. After extracting aqueous extracts by hot method, silver nanoparticles were synthesized by the biological method. Green synthesized silver nanoparticles were analyzed by UV–Vis spectroscopy, XRD, FTIR, and FESEM-EDAX. The antibacterial effect was evaluated by diffusion method in agar and determination of minimum growth inhibitory and lethal concentration (MIC and MBC) by dilution method in liquid culture medium. Results Based on the results of UV–Vis spectroscopy, silver nanoparticles synthesized from N. sessilifolia and S. hydrangea had distinct absorption peaks at wavelengths of 407 to 424 nm and 414 to 415 nm, respectively. The crystalline nature of these synthetic silver nanoparticles was confirmed by XRD. FESEM analysis showed that the size of biosynthesized silver nanoparticles from N. sessilifolia and S. hydrangea extracts were 10–50 nm and 10–80 nm, respectively, and were cubic. The results of diffusion in agar showed that the largest diameter of the growth inhibition zone belonging to the synthetic silver nanoparticles from both extracts of N. sessilifolia (~ 26.00 mm) and S. hydrangea (~ 23.50 mm) was against Gram-positive bacteria Staphylococcus aureus. The most vigorous killing activity by synthetic silver nanoparticles from N. sessilifolia extract was against Klebsiella pneumoniae with a value of 250 μg/mL, two times stronger than rifampin. Conclusion Therefore, the studied extracts can be suitable options for fast and safe green synthesis of silver nanoparticles effective against some bacterial strains. These synthetic silver nanoparticles can be used as possible options and have strong potential for the production of natural antibiotics.

Studies have shown an inverse correlation among age-related illnesses like coronary heart disease and cancer and intake of fruit and vegetable. Given the probable health benefits of natural antioxidants from plants, research on them has increased. Dactylorhiza osmanica is consumed as a food and traditional medicine plant in some regions of Turkey, so evaluation of the biological ability of this species is important. In this study, the amount of phenolic content (LC-HRMS), antioxidant activities and enzyme inhibitory properties of an endemic plant, D. osmanica, were investigated. The antioxidant capacities of an ethanol extract of D. osmanica aerial parts (EDOA) and roots (EDOR) were evaluated with various antioxidant methods. Additionally, the enzyme inhibitory effects of EDOA and EDOR were examined against acetylcholinesterase (AChE), α-glycosidase, and α-amylase enzymes, which are associated with common and global Alzheimer’s disease and diabetes mellitus. The IC50 values of EDOA against the enzymes were found to be 1.809, 1.098, and 0.726 mg/mL, respectively; and the IC50 values of EDOR against the enzymes were found to be 2.466, 0.442, and 0.415 mg/mL, respectively. Additionally, LC-HRMS analyses revealed p-Coumaric acid as the most plentiful phenolic in both EDOA (541.49 mg/g) and EDOR (559.22 mg/g). Furthermore, the molecular docking interaction of p-coumaric acid, quercitrin, and vanillic acid, which are the most plentiful phenolic compounds in the extracts, with AChE, α-glucosidase, and α-amylase, were evaluated using AutoDock Vina software. The rich phenolic content and the effective antioxidant ability and enzyme inhibition potentials of EDOA and EDOR may support the plant’s widespread food and traditional medicinal uses.
