Background. Phenolic compounds were extracted and isolated from S. undulata roots. Methods. Sample of roots from E. hirta was tested for phenolic compounds, and in vitro antioxidant activity by diphenyl-1-picrylhydrazyl (DPPH) assay, ABTS, FRAP and reducing power was measured using cyanoferrate method. Results. The methanolic fraction exhibited the highest total phenol content (6.12 ±0.11 mg AGE/g DW). On the other hand, the highest fl avonoids concentration was observed in ethyl acetate fraction (2.90 ±0.05 mg CE/g DW) in addition to anthocyanins (28.56 ±3.96 mg/l). Besides, the highest level of tannins content was measured in the polar aprotic solvent ethyl acetate extract (3.25 ±0.06 mg CE/g DW). The different extracts of S. undulata were evaluated for their radical scavenging activities by means of the DPPH assay. The strongest scavenging activity was observed in methanolic fraction scavenged radicals effectively with IC 50 values of 0.14 ±0.02 mg/ml. Similarly, the potassium ferricyanide reduction (FRAP) and ABTS•+ of methanol extract. On the other hand, the total reducing power of ethyl acetate extract was found higher than of other extracts. This paper presents the application of the design-of experiment method for optimizing the extraction of phenolic content using methanol solvent. The resulting regression model has shown that the effect of temperature is not statistically signifi cant (with >95% certainty), while that of agitation speed is. The two main effects are contributed by the solvent concentration and the maceration period. Conclusion. Our results clearly showed that the extraction of phenolic compounds and their antioxidant capacity is signifi cantly affected by solvent combinations. S. undulata presented the highest total phenolic content, total fl avonoids content and antioxidant capacity values. ÖzetBu çalışmada; P. europea'nın besinsel içerikleri, antioksidan ve antimikrobiyal etkileri araştırıldı. Şeker, flavonoid, vitamin ve mineral element düzeylerinin değişebildiği gözlendi. In vitro koşullarda, DPPH radikalini temizleme aktivitesinin 10 µg/µL'de %83.62, FeCl grubunun K grubuna göre yüksek ve metil esterlerinin ise korunmadığı belirlendi. P. europea'nın antimikrobiyal aktivitesinin ise; maya ve dermofit funguslardan ziyade, bakteri gruplarında daha etkili olduğu saptandı.Anahtar kelimeler: Antimikrobiyal aktivite, antioksidan, besinsel içerik, P. europea AbstractIn this study, nutritive value, antioxidant and antimicrobial activitiy of P. europea was determined. It was observed that sugar, flavonoid, vitamin and element levels varied. It was understood that under in vitro conditions the DPPH activities had a level of 83.62% at 10 µg/µL, the FeCL group was higher than the K group, and the methyl esters were not preserved as well. It was also determined that the antimicrobial activity of P. europea is more efficent over bacterial groups then yeast and dermatophyte fungi.

The principles of good agricultural and horticultural practice, considering both environmental protection and high yielding of plants, require modern cultivation methods. In modern agriculture, it is possible to use biostimulants that protect the soil against degradation and plants against phytopathogens and stress. The purpose of 3-year field and laboratory studies was to determine the effect of Trichoderma harzianum T-22 and other biostimulants on the health status of scorzonera (Scorzonera hispanica L.) plants and microorganism populations in the rhizosphere of this plant. For this purpose, Biosept Active (a.s.—grapefruit extract), Timorex Gold 24 EC (based on tea tree oil), Trianum P (spores of Trichoderma harzianum Rifai T-22) and Zaprawa Nasienna T 75 DS/WS fungicide (a.s.—tiuram) were applied for the pre-sowing seed dressing of scorzonera cv. “Duplex”. The number of seedlings and the health status of scorzonera plants were determined during three growing seasons. In each year of the study, mycological analysis of seedling roots and roots after scorzonera harvest was conducted to establish the quantitative and qualitative composition of fungi colonizing these parts. Moreover, microbiological analyses of scorzonera rhzisphere soil were conducted and served as the basis to determine the total population of fungi and bacteria (including Pseudomonas sp. and Bacillus sp.). Antagonistic activity of rhizosphere bacteria Pseudomonas sp., Bacillus sp. and fungi was determined based on laboratory tests on selected scorzonera soil-borne fungal pathogens (Alternaria scorzonerae, Fusarium culmorum, Fusarium oxysporum, and Rhizoctonia solani). The experiments showed that Trianum P most effectively protected the roots of scorzonera against infection by Alternaria alternata, A. scorzonerae, Neocosmospora solani, Fusarium spp., Sclerotinia sclerotiorum, Rhizoctonia solani, and Botrytis cinerea. The rhizosphere population of Bacillus sp. and Pseudomonas sp. in the treatments with Trianum P or Zaprawa Nasienna T 75 DS/WS was larger than in the other experimental treatments. A reverse relationship was observed in the population of rhizosphere fungi. The application of grapefruit extract, tea tree oil and Trichoderma harzianum T-22 increased antagonistic activity of Pseudomonas sp., Bacillus sp. and selected saprotrophic fungi against soil-borne fungal pathogens, especially Alternaria sp., Rhizoctonia sp., and Fusarium sp. In summary, Biosept Active, Timorex Gold 24 EC and Trianum P can be recommended as plant biostimulants in Scorzonera hispanica cultivation.
