Considering the challenge to control Candida -associated denture stomatitis, the search for antifungal substances derived from natural sources has become a trend in the literature. In this study the following effects of Chenopodium ambrosioides extract (CAE) were investigated: action against biofilms of Candida albicans , its cytotoxic potential, and changes caused in acrylic resin. The CAE was characterized by High Performance Liquid Chromatography (HPLC). The susceptibility of C. albicans to CAE was investigated by Minimum Inhibitory Concentration and Minimum Fungicidal Concentration (MIC and MFC) tests. Acrylic resin disks were fabricated, and C. albicans biofilms were developed on these for 48 h. Afterward the disks were immersed for 10 min in: PBS (Negative Control); 1% Sodium Hypochlorite (1% SH, Positive Control) or CAE at MIC or 5xMIC. The biofilms were investigated relative to counts and metabolic activity. The cytotoxic potential in keratinocytes and fibroblasts was verified by MTT test. Change in color and roughness of the acrylic resin was analyzed after 28 days of immersion in CAE. The data were analyzed by the ANOVA considering a 5% level of significance. The main compounds detected by HPLC were kaempferol and quercetin. Both MIC and MFC obtained the value of 0.25 mg/mL. The MIC was sufficient to significantly reduce the counts and activity of the biofilm cells ( p < 0.0001), while 5xMIC resulted in almost complete eradication, similar to 1% SH. Keratinocytes and fibroblasts exposed to the MIC and 5xMIC presented cell viability similar to that of the Control Group ( p > 0.05). No important changes in acrylic resin color and roughness were detected, even after 28 days. It could be concluded that the immersion of acrylic resin in C. ambrosioides extract in its minimum inhibitory concentration was effective for the reduction of C. albicans biofilms without any evidence of cytotoxic effects or changes in roughness and color of this substrate.

O estudo tem por objetivo descrever os resultados obtidos dos parâmetros de qualidade farmacognóstico, prospecção fitoquímica e estudos in silico de monoterpenos isolados de Dysphania ambrosioides. Utilizou-se folhas e caules que foram secas e pulverizadas, os pós foram submetidos a testes farmacognóstico. Por meio de maceração com etanol obteve-se os extratos de caule (EEC) e de folhas (EEF), seguido de prospecção fitoquímica através de cromatografia em camada delgada. Para os estudos in silico selecionou-se os terpenos (1S,2S,3R,4S)-1-metil-4-(propan-2-il)-ciclohexano-1,2,3,4-tetrol (A); 1,2,3,4-tetrahidroxi-p-metano (B); (1R,2S)-3-pmeten-1,2-diol (C); (1R,4S)-p-met-2-en-1-ol (D); 1,4-dihidroxip-met-2-eno (E); 1-metil-4β-isopropil-1-ciclohexeno-4α, 5α, 6α-triol (F); Ascaridol (G), onde utilizou-se os programas Marvin JS, PreADMET, PASS online, Molinspiration Online, para avaliação dos aspectos físico-químicos, farmacocinéticos, toxicológicos e predição de atividade. Os pós foram classificados como grossos, com teor de umidade dentro dos parâmetros recomendados, sendo ausente as saponinas, pH neutro e baixa densidade. Na prospecção fitoquímica foram detectados triterpenos e esteroides, saponinas e heterosídeos flavônicos. Todos os compostos seguem a regra de Lipinski; foram bem absorvidos no trato gastrointestinal; C, D, E e G ligam-se fortemente às proteínas plasmáticas; C e D distribuíram amplamente no sistema nervoso central; D e E não foram metabolizadas pelo CYP e todas inibiram CYP; A e B não foram mutagênicas e F e G são carcinogênicas. As drogas vegetais apresentaram boa qualidade, sendo os terpenos presentes nestas amostras. Os estudos in silico apontam que as moléculas as moléculas C, F e G são promissoras para atividade anti-inflamatória, antineoplásica e antiprotozoária.

Dysphania ambrosioides (L.) Mosyakin and Clemants is a medicinal plant that has traditionally been used to cure a range of diseases. There has been no thorough investigation of the potential toxicity of this plant. The objective of this study is to assess the acute and subacute toxicity of D. ambrosioides hydroethanolic extract (DAHE), as well as it alkaloids composition, utilizing LC-MS/MS analysis. An in silico approach was applied to determine pharmacokinetic parameters and to predict the toxicity of D. ambrosioides identified alkaloids. A 14-day treatment with a single oral dose of 1–7 g/kg was carried out to investigate acute toxicity. DAHE was given orally at dosages of 5, 50, and 500 mg/kg for 15 days in the subacute toxicity investigation, and body weight and biochemical parameters were evaluated. Livers, kidneys, lungs, and heart were examined histologically. Chromatographic investigation revealed the existence of nine alkaloids, with N-formylnorgalanthamine being the most prevalent. The oral LD50 value of DAHE was found to be 5000 mg/kg in an acute toxicity study. No variations were observed with respect to food intake, water consumption, mortality, or body and organ weight in the subacute toxicity study. On the other hand, DAHE (500 mg/kg) significantly enhanced alanineaminotransferase, aspartate aminotransferase, and urea. Liver and kidney histological examinations revealed modest infiltration of hepatocyte trabeculae by inflammatory cells in the liver and slight alteration in the kidney histoarchitecture. According to our findings, DAHE exhibits low to moderate toxicity.
