Physiological regulation of cardiac contractility by endogenous reactive oxygen species

Abstract: Increased production of reactive oxygen species (ROS) has been linked to the pathogenesis of congestive heart failure. However, emerging evidence suggests the involvement of ROS in the regulation of various physiological cellular processes in the myocardium. In this review, we summarize the latest findings regarding the role of ROS in the acute regulation of cardiac contractility. We discuss ROS-dependent modulation of the inotropic responses to G protein-coupled receptor agonists (e.g. β-adrenergic receptor a… Show more

“…Well is known that NADPH oxidase generates superoxide anion, which dismutates to form H 2 O 2 , the main variant of reactive oxygen species in cells under physiological conditions (Miller et al, 2010;Perjes et al, 2012). So we examined the assumption that reactive oxygen species could be derived from the membrane-bound NADPH-oxidase.…”

“…The major sources of reactive oxygen species in cardiomyocytes are membrane-bound NADPHoxidases and mitochondrial electron-transition complexes (Perjes et al, 2012). Recently, using transgenic mice with β 2 -adrenoceptor overexpression it has been shown that chronic stimulation of cardiac β 2 -adrenoceptors leads to an increase in reactive oxygen species production via activation of NADPH-oxidase (Xu et al, 2011).…”

“…ROS play an important role in the regulation of normal cardiac function by modulating the function of various kinases, phosphatases, ion channels, transporters and myofibrillar proteins [94]. However, excessive ROS production has been implicated in the development of cardiac dysfunction in different pathological conditions, including HF and DM, and the evidence suggests that oxidative PTM (OPTM) of Ca 2+ handling proteins, such as SERCA2a and RyR2, may be involved [95][96][97].…”

“…Az antioxidáns vegyületekkel folytatott klinikai tanulmányok ez idáig csalódást okoztak, a ROS-termelődés csökkentése nem javította a szív pumpafunkcióját a szívelégtelen betegekben általánosan [32]. A ROS jelátviteli szerepének felismerése a szív fi ziológiás folyamatainak szabályozásában [11], illetve a stresszhatásokra adott adaptációs válaszban [20] magyarázatot adhat az antioxidáns vegyületekkel folytatott kezelések sikertelenségére.
