Physiological and Regulatory Effects of Controlled Overproduction of Five Cold Shock Proteins of Lactococcus lactis MG1363

Abstract: The physiological and regulatory effects of overproduction of five cold shock proteins (CSPs) of Lactococcus lactis were studied. CspB, CspD, and CspE could be overproduced at high levels (up to 19% of the total protein), whereas for CspA and CspC limited overproduction (0.3 to 0.5% of the total protein) was obtained. Northern blot analysis revealed low abundance of the cspC transcript, indicating that the stability of cspC mRNA is low. The limited overproduction of CspA is likely to be caused by low stability… Show more

“…CSPs have been shown to affect the synthesis of other cytosolic proteins at 37°C and after cold shock (5,6,18,22,33). gested that CSPs function as RNA chaperones (17) by binding to newly synthesized RNA and by keeping RNA in a linear form (6).…”

“…Previous studies on the chromosomal CspC and CspD homologues have illustrated the physiological response mediated by these cold shock proteins; expression of CspD was enhanced by cold shock and resulted in increased survival at low temperatures, whereas only a modest increase in CspC expression accompanied cold shock, which was found to directly alter the levels of other cold shock proteins (71)(72)(73).…”

“…The supernatant was removed and pellets were suspended in 1% (wt/vol) osmium tetraoxide for 1 hr. The cells were then harvested through a graded ethanol series (30,50,70, 90 and 100%) and infiltrated with Araldite (Sigma). Resin blocks were polymerized at 60 C for 36 hr.…”
