Physiological and Pathological Remodeling of Cerebral Microvessels

Abstract: There is growing evidence that the remodeling of cerebral microvessels plays an important role in plastic changes in the brain associated with development, experience, learning, and memory consolidation. At the same time, abnormal neoangiogenesis, and deregulated regulation of microvascular regression, or pruning, could contribute to the pathogenesis of neurodevelopmental diseases, stroke, and neurodegeneration. Aberrant remodeling of microvesselsis associated with blood–brain barrier breakdown, development of… Show more

“…The assessment of neurovascular dysfunction is of particular interest as it plays a crucial role in the onset and progression of the neurodegenerative process associated with the accumulation of β-amyloid peptide in brain neurons and cerebral vessel walls [ 85 , 86 ]. Special attention is given to the dysfunction of the blood–brain barrier, reduced cerebral blood flow, and impaired vascular clearance of beta-amyloid from the brain into the glymphatic system and meningeal lymphatic vessels [ 87 ].…”

“…Endothelial cells take part in the establishment of a new microvascular network either in embryogenesis (developmental vasculogenesis and angiogenesis) or in the adult brain (experience-and damage-driven angiogenesis). These mechanisms are based on the activity of endothelial progenitor cells, migrating non-proliferating tip-type endothelial cells, and proliferating stalk-type endothelial cells in the tissue [153]. Uptake of extracellular glucose for energy production is indispensable for angiogenesis and barriergenesis; thereby, blocking glucose transport in brain endothelial cells disrupts postnatal angiogenesis and the establishment of the barrier [154].…”

“…Abnormal angiogenesis is involved in the pathogenesis of Moyamoya disease, which is often accompanied by cognitive and developmental delays or disability [127]. Aberrant angiogenesis and remodeling of brain microvessels is associated with disorganization of the BBB, neuroinflammation, and dysfunction of the neurovascular unit, resulting in neurological deficits [128,129]. In contrast, angiogenesis has curative roles in the recovery from ischemic stroke and depression [130,131].…”
