This study examined the role of seed ageing in the control of anti-nutritional factors in cowpea (Vigna unguiculata L.). In differently aged seeds of three cultivars of V. unguiculata (V240, V78 and V585), germination ability and vigour were studied. Effort was also made to assay trypsin inhibitor, phenol and phytic acid, perform protein profiling in these seeds. High vigour lots (V240 and V585) registered maximum increases in germination of aged seeds. The contents of nutritional factors such as total protein and carbohydrate declined with decrease in seed vigour lots. Antinutritional factors such as phytic acid, phenolic content and trypsin inhibitor activity decreased and varied in low and high vigour seed lots. Polypeptide banding pattern significantly varied in the high, medium and low vigour seeds. Notably, proteins with the highest relative mobility of 0.98 and lowest molecular weight of 11.5 kDa and lowest relative mobility of 0.17 and highest molecular weight of 102.0 kDa were observed in all the vigour lots. Results implied the decline in vigour of V. unguiculata seeds under conditions of controlled ageing can be related to the decline in content of major nutritional factors (total carbohydrates and proteins) required for the growing embryo during seed germination. Additionally, decreases in the contents of anti-nutritional factors phytic acid and phenols, and the activity of trypsin inhibitor in particular are connected with the decrease in seed vigour irrespective of V. unguiculata cultivars. The use of short duration controlled ageing technique can, at least partially, reduce the negative effects of anti-nutritional factors, and eventually improve the nutritional quality of V. unguiculata seeds.

Chemical analysis and mineral composition of twenty accessions of grain and leaf Amaranth (Amaranthus L.) collected from different agro-ecological zones of Central Malawi were conducted according to the standards of Association of Official Analytical Chemistry (AOAC). Analysis of variance (ANOVA) and means were separated using least significance difference (P ≤ 0.05) in Gen Stat version 15. The analyses for grain Amaranth showed that moisture content ranges from 10.69 to 12.22% while ash content varied from 4.4 to 8.7%. Elemental analyses in mg/100 grams on dry weight basis indicated that the grain had calcium (78.3 to 1004.6), iron (3.61 to 22.51), magnesium (44.31 to 97.38), potassium (267.8 to 473.6) and zinc (0.53 to 1.20). The mean differences for leaf chemical analyses were highly significant (p < 0.001) with crude protein ranging from 13.37 to 23.27%; ash (14.08 to 19.95%) and Vitamin C (30.3 to 117.79 mg/100 g) while the mean mineral leaf analyses in mg/100 grams ranged from 14.84 to 31.17 for iron, 1.03 to 3.46 for zinc, 1512 to 2381 for calcium, 1320 to 1677 for potassium and 383.4 to 513.9 for magnesium. Generally the accessions from mid altitude area of Lilongwe showed highest values for both grain and leaf mineral analyses while accessions from the high altitude showed lower values. The results of this study provide evidence that local Amaranthus genotypes have appreciable amount of nutrients, minerals and vitamins important to meet dietary requirements of rural and urban communities in Malawi.

Two experiments were conducted to evaluate ageing-induced genetic changes and to establish physiological thresholds for loss of genetic integrity during ageing or storage of rice seeds. In the first experiment, seeds of 10 varieties of rice were subjected to artificial ageing in chambers conditioned to 55°C and 72±2% RH for 72 hours. In the second experiment, seeds of 4 varieties of rice stored in the NACGRAB gene bank, Nigeria in 2011 at 5± 4°C were compared with seeds of the same accessions freshly harvested in 2013. Data were collected on seed germination and seedling length to estimate the seed vigour index. Genetic changes during the ageing were evaluated by SSR markers using a Direct™ PCR kit. Genetic distance indices were computed using PAST™ software and percentage genetic integrity was estimated from the genetic distance matrices. At 72 hours of artificial ageing, seed germination percentage declined to 54.2% and vigour index 0.8 coinciding with the lowest estimate of genetic integrity of 99.5%. The decline in percentage genetic integrity during the artificial ageing indicated a systematic ageing-induced genetic alteration. SSR primer RM178 revealed variations that suggest losses of alleles in the course of ageing for 2 accessions at between 24 hours in WITA4 and 48 to 51 hours in CG14. In the gene bank storage trial, germination of seeds after one and two years of storage was above 80% and there were no significant differences among the accessions. SSR profiles for all the accessions were also similar. The result partly corroborates the artificial ageing data. This implies that seed viability benchmark of 54% is recommended for regeneration of stored rice seeds in order to maintain optimum genetic integrity during storage.
