Antioxidants play an important role in adapting plants to abiotic stress by detoxifying reactive oxygen species (ROS). Involvement of antioxidant enzymes in abiotic stress tolerance of highly stress-tolerant quinoa was studied in a climatic chamber at 6 mOsm (milliosmolar) ionic (300 mM NaCl) and non-ionic (600 mM mannitol) salts combined with increasing levels of potassium K1 and K2 (6, 12 mM), respectively. Fifteen days of salt treatment (both ionic and nonionic) decreased plant growth (shoot and root fresh weight), stomatal conductance and chlorophyll content index. Furthermore, both forms of salt stress increased the activities of superoxide dismutase, catalase, ascorbate peroxidase and peroxidase up to 2.33-, 3.98-, 4.78-and 5.55-folds, respectively, compared to no salt treatment, whereas membrane stability index decreased corresponding to increase in lipid peroxidation (malondialdehyde), with salt treatments compared to non-stressed plants. However, no significant effect of potassium and salt treatments has been noticed on the maximal photochemical efficiency of PSII. The results suggested that enhanced antioxidant enzymes activity under salt stress could be one of the factors responsible for abiotic stress tolerance in quinoa.

Protein extraction for two-dimensional electrophoresis from tissues of recalcitrant species is quite problematic and challenging due to the low protein content and high abundance of contaminants. Proteomics in Shorea robusta is scarcely conducted due to the lack of a suitable protein preparation procedure. To establish an effective protein extraction protocol suitable for two-dimensional electrophoresis in Shorea robusta, four procedures (borate buffer/trichloroacetic acid extraction, organic solvent/trichloroacetic acid precipitation, sucrose/Tris/phenol, and organic solvent/phenol/sodium dodecyl sulfate) were evaluated. Following these, proteins were isolated from mature leaves and were analyzed for proteomics, and also for potential contaminants, widely reported to hinder proteomics. The borate buffer/trichloroacetic acid extraction had the lowest protein yield and did not result in any banding even in one-dimensional electrophoresis. In contrast, organic solvent/phenol/sodium dodecyl sulfate extraction allowed the highest protein yield. Moreover, during proteomics, organic solvent/phenol/sodium dodecyl sulfate extracted protein resolved the maximum number (144) of spots. Further, when proteins were evaluated for contaminants, significant (77-95%) reductions in the nucleic acids, phenol, and sugars were discernible with refinement in extraction procedure. Accumulated data suggested that the organic solvent/phenol/sodium dodecyl sulfate extraction was the most effective protocol for protein isolation for proteomics of Shorea robusta and can be used for plants that have a similar set of contaminants.

Lipid peroxidation and seed emergency in progenies of the yellow passion fruit plant

-The objective was to evaluate the percentage of emergency plantlets and lipid peroxidation in seeds of 29 half-sib progenies of yellow passion fruit (Passiflora edulis Sims.) after 24 months under storage. The experimental design was completely randomized, with four replications of 50 seeds each, from which the treatments were the progenies (1-29). The evaluation of the percent plantlet emergency was accomplished at 14 and 28 days after sowing. The lipid peroxidation of the seeds was expressed as malondialdehyde (MDA) content that was determined by the TBARS method. Approximately 21% of those half-sib progenies maintained the viability of their seeds for twenty-four months under storage. The results point out a remarkable genetic variability for vigor and emergency of the yellow passion fruit plantlets, with occurrence of individuals with high and other ones with low capacity to maintaining the physiologic quality of their seeds after storage. O delineamento experimental foi o inteiramente casualizado, com quatro repetições de 50 sementes cada, a partir do qual os tratamentos foram as progênies (1, 2, 3, 4, 5, 6, 7, 8, 9, 10, 11, 12, 13, 14, 15, 16, 17, 18, 19, 20, 21, 22, 23, 24, 25, 26, 27, 28 e 29). A avaliação da porcentagem de emergência das plântulas foi realizada aos 14 e 28 dias após a semeadua. A peroxidação lipídica das sementes foi expressa como conteúdo de malondialdeído (MDA), que foi determinado pelo método de TBARS. Aproximadamente 21% das progênies de meios-irmãos mantiveram a viabilidade de suas sementes para 24 meses de armazenamento. Os resultados apontam para uma variabilidade genética quanto ao vigor e á emergência de plântulas de maracujazeiro-amarelo, com ocorrência de indivíduos com alta e outros com baixa capacidade de manter a qualidade fisiológica de suas sementes após o armazenamento.
