Fungal chitosan was extracted from Aspergillus niger mycelia. The produced chitosan was characterized with deacetylation degree of 89.2%, a molecular weight of 2.4 × 10 4 Da, and 96.0% solubility in 1% acetic acid solution. The antibacterial activity of fungal chitosan was evaluated against two foodborne pathogens, that is, Salmonella typhimurium and Staphylococcus aureus, using the established antibacterial assays, for example, zone of growth inhibition and agar plat count tests, and using 2,3,5,-triphenyltetrazolium chloride (TTC) as chromogenic marker for qualitative and quantitative determining of antibacterial potentiality. The TTC (0.5% w/v) was added, at concentration of 10%, to cultured broth, containing chitosan with different concentrations then the formed formazan was separated. The formation of red formazan could be considered as a qualitative indication for antibacterial activity, whereas the measurement of color intensity for the resuspended red formazan, using spectrophotometer at 480 nm, provided a quantitative evidence for the strength of the used antibacterial agent. Regarding the rapidity, technical simplicity, and cost-effectiveness, TTC assay could be recommended as an efficient alternative method for qualitative and quantitative determination of chitosan antibacterial activity and could be suggested for general evaluation of antibacterial agents.

Two hundred and six bacterial isolates were obtained from leaf, flower and stem of three healthy plants, Euphorbia pulcherrima Willd, Pyrethrum cinerariifolium Trev. and Heracleum candicans Wall. The nematicidal activity experiment showed that a total of 92 isolates displayed activity against Caenorhabditis elegans (Maupas) Dougherty, and 70 isolates resulted active against Bursaphelenchus xylophilus (Steiner & Buhrer) Nickle. Strain LCB-3 exhibited the strongest nematicidal activity against both two nematodes. According to the 16S rDNA, the strain LCB-3 was identified as Brevundimonas diminuta. Based on bioassay-guided fractionation, a nematicidal metabolite (R)-(-)-2-ethylhexan-1-ol was obtained from LCB-3. The median lethal concentrations (LC 50 ) of the compound were 542.0 mg l -1 against C. elegans and 168.1 mg l -1 against B. xylophilus 48 h after treatment.

In this study, we investigated the interface contacting inhibition behaviors of chitosan against bacterial in the dispersing state. For that purpose, chitosan microspheres (CMs) in the dispersing state was prepared by the emulsification cross-linking method. The CMs had smooth surface and spherical shape with the diameter of about 124 mm. They were stable after sterilization at 121uC and 150 kPa for 20 min. The CMs had similar antibacterial activity to that of chitosan in the solution form. Their antibacterial activities increased with the increase of the CM concentration, while decreased with the increase of pH of the system. It was found that the CMs with the degree of deacetylation (DD) of 63.6% exhibited the highest antibacterial activity, while the CMs with the DD of 83.7% exerted the lowest antibacterial activity among the three tested samples.
