Physical Activity Interferes with the Immunomodulatory Effect of the Antineoplastic Drug NSC631570

Abstract: NSC 631570 causes M1 (N1) shift of phagocytes after in vivo introduction. Moderate physical exercise exerts a negative effect on the immunomodulatory action of NSC 631570 by abrogating M1 (N1) shift of circulating phagocytes. One of the reasons for such an effect could be an increase in PPAR-γ expression by phagocytes.

“…Neutrophils produce active forms of oxygen, which contributes to the development of inflammation (Fernández-Sánchez et al, 2011;Lutai et al, 2016). The functional polarization of phagocytes (neutrophils and monocytes), characterized by the ratio of the production of reactive forms of oxygen and phagocytic activity, depends on many factors, in particular, the period of the day, physical activity, the state of lipid metabolism (Skivka et al, 2015a(Skivka et al, , 2015bRudyk et al, 2016Rudyk et al, , 2018. There are data on dependence of functional status of neutrophils on thyroid status parameters (Mezosi et al, 2005;Marino et al, 2006;Mathison, 2006;Kurjane et al, 2012).…”

“…Granulocytes or monocytes were gated according to forward and side scatters. Phorbol 12-myristate 13-acetate (PMA) (Sigma-Aldrich) was used to evaluate nonspecific reactivity reserve in phagocytes (Shapiro et al 2011 ; Skivka et al 2015 ). Reactivity reserve was characterized by the modulation coefficient (MC) that was calculated by the following formula: where S is the ROS value in probes stimulated with PMA in vitro, and B is ROS value in unstimulated probes (basal value).…”

“…The number of monocytes in peripheral blood does not change significantly with age, age-related changes are mainly related to macrophage activity, however, there is evidence of phagocytosis inhibition in monocytes and the production of reactive oxygen (Plowden et al, 2004;Hearps et al, 2012;Metcalf et al, 2015). In general, when exposed to extreme exogenous factors, phagocytic activity is indicated among the sensitive factors of innate immunity (Skivka et al, 2015a;Boieva et al, 2018). The age-related changes in the number and function of NK cells, which are considered to be short-lived effector cells, are quite controversial.…”
