Phylum-Spanning Neuropeptide GPCR Identification and Prioritization: Shaping Drug Target Discovery Pipelines for Nematode Parasite Control

Abstract: Nematode parasites undermine human health and global food security. The frontline anthelmintic portfolio used to treat parasitic nematodes is threatened by the escalation of anthelmintic resistance, resulting in a demand for new drug targets for parasite control. Nematode neuropeptide signalling pathways represent an attractive source of novel drug targets which currently remain unexploited. The complexity of the nematode neuropeptidergic system challenges the discovery of new targets for parasite control, how… Show more

“…Orthologs of known anthelmintic target genes, as previously identified in C. elegans [ 23 ], were retrieved from WormBase Parasite. Additional targets of interest were taken from [ 24 ]. With the H. bakeri locus tags, expression of each target was examined.…”

“…Only two of these receptors, NPFR-1 and NPFR-2 encoded by the npr-11 and npr-12 genes, cluster together with invertebrate NPF receptors in bilaterian-wide phylogenetic analyses and thus represent orthologs of the NPY/F receptor family ( Beets et al, 2023 ; Jékely, 2013 ; Mirabeau and Joly, 2013 ). Both receptors are also well conserved in the nematode phylum ( Atkinson et al, 2021 ; Elphick and Mirabeau, 2014 ). By contrast, the family of sNPF receptors expanded in nematodes and includes the receptors NPR-1 to NPR-7, NPR-10, and NPR-13 in C. elegans ( Beets et al, 2023 ; Jékely, 2013 ; Mirabeau and Joly, 2013 ); their functions have been reviewed elsewhere ( Fadda et al, 2019 ).…”
