Phylogeny of the Glomerales and Diversisporales (Fungi: Glomeromycota) from actin and elongation factor 1-alpha sequences

Abstract: The arbuscular mycorrhizal (AM) fungi have been elevated to the phylum Glomeromycota based on a ribosomal gene phylogeny. In order to test this phylogeny, we amplified and sequenced small subunit ribosomal RNA (SSUrRNA), actin and elongation factor 1 (EF1)-alpha gene fragments from single spores of Acaulospora laevis, Glomus caledonium, Gigaspora margarita, and Scutellospora dipurpurescens. Sequence variation within and among spores of an isolate was low except for SSUrRNA in S. dipurpurescens, and the actin a… Show more

“…Specific 2-aa deletions (Tanabe et al, 2002) are boxed. We confirmed that this deletion is also present in the recently published sequence of the Glomales (Helgason et al, 2003). In addition to sequences used in Tanabe et al (2002Tanabe et al ( , 2004, sequences of Ichthyophonus irregularis (GenBank acc.…”

“…From these results, Schüßler et al (2001) revised the higher-level taxonomy of the Glomales: the Glomales (sensu Morton and Benny, 1990) was elevated to the new phylum Glomeromycota, and the four orders, the Archaeosporales, Diversisporales, Glomerales, and Paraglomales were revised or newly erected within the Glomeromycota. However, subsequent analyses using EF-1a and actin have not supported a phylogenetic affinity of the Glomales with higher fungi (Helgason et al, 2003). Results of an a-tubulin analysis further complicated the picture by recovering a monophyletic grouping of the Glomales and Chytridiomycota (Corradi et al, 2004).…”

“…Taxonomy of AMF is presently changing dramatically, and as a consequence, several new phylogenetic units have been established (Helgason et al 2003;Schüβler et al 2001b;Schwarzott et al 2001). For this reason we determined the approximate phylogenetic position of the myco-heterotrophic plant mycobionts by comparison with phylogenetic treatments, representing the basis of the new AMF taxonomy (Schüβler et al 2001b;Schwarzott et al 2001).…”

“…Some of them include DNA/DNA hybridization, G+C content analysis or PCR based methods, including those that detect polymorphisms in the genome, like RAPDs and AFLPs, or in certain genomic regions or genes, like RFLPs (for ribosomal RNA) or direct sequencing of these and other genomic regions (ribosomal DNA, actin gene, transcription elongation factor, beta tubulin gene, Ras protein, chitin synthase, etc.) (Bruns et al, 1991;Mehmann et al, 1994;Thon and Royse, 1999;Helgason et al, 2003).…”
