Lima bean plants showing mosaic and leaf distortion symptoms, similar to those induced by viruses, were observed in several production areas in the states of Ceará and Piauí, Northeastern Brazil. The aim of this study was to identify RNA genome viruses that infect lima bean. Field research was conducted from 2017 to 2018. Fifty-five symptomatic samples were randomly collected from seven properties in five municipalities and tested by RT-PCR and DNA sequencing with specific or universal primers for two viruses and two virus groups. Four virus species were identified: cucumber mosaic virus (CMV), cowpea mild mottle virus (CPMMV), cowpea aphid-borne mosaic virus (CABMV) and cowpea severe mosaic virus (CPSMV), which had infection levels of 21.8, 52.7, 47.2, and 1.8%, respectively, as well as double and triple infections. The CMV isolates belonged to subgroup IA. The CPMMV isolates had high nucleotide identity with CPMMV isolates from Brazil, USA and Mexico. The CABMV isolates showed moderate nucleotide identity with Brazilian isolates. Only one sample was infected with CPSMV. This is the first record of CPMMV and CPSMV naturally infecting lima bean. Approaches to virus control are discussed.

Passion fruit (Passiflora edulis) is a fruit species of great relevance for Brazilian economy. However, it is highly susceptible to the Cowpea aphid-borne mosaic virus (CABMV). The species P.setacea, on the other hand, is resistant to the disease. The present study aimed to identify CABMV-resistant P. setacea genotypes for the introgression of genes into sour passion fruit genetic breeding programs. The seeds of passion fruit genotypes were germinated in vitro in MS culture medium. The seedlings were acclimatized in a growth chamber at the temperature of 27 ± 2ºC, photoperiod of 16:8 hours and 80% of relative humidity. Thirty plants of each genotype were mechanically inoculated with extract prepared from leaves collected from passion fruit plants with symptoms of CABMV for the assessment of resistance to CABMV. The severity of leaf symptoms was evaluated by means of a grading scale of visual signs. After the visual evaluation and identification of the asymptomatic genotypes of P. setacea, the PTA-ELISA test was carried out for 30 selected genotypes. According to the visual evaluation, all P. setacea genotypes were highly resistant to CABMV, while the P. edulis genotypes were highly susceptible. Out of the 30 genotypes selected, only PsRJ 4 was considered susceptible by PTA-ELISA. The other genotypes of P. setacea were considered resistant and present great potential for use in passion fruit genetic breeding programs.

BackgroundPassion fruit woodiness may be caused by Cowpea aphid-borne mosaic virus (CABMV) and is currently the major passion fruit disease in Brazil. To assess the virus-vector-host interactions, a newly introduced golden passion fruit plantation located in eastern region of São Paulo State, Brazil, was monitored.MethodsDissemination of CABMV was determined analyzing golden passion fruit plants monthly for 18 months by PTA-ELISA. Seasonality and aphid fauna diversity was determined by identification of the captured species using yellow sticky, yellow water-pan and green tile traps. Population composition of the aphid species was determined using the descriptive index of occurrence, dominance and general classification and overlap of species in the R program.ResultsAnalyses of species grouping afforded to recognize 14 aphid species. The genus Aphis represented 55.42 % of the species captured. Aphid species formed two distinct clusters, one of which was characterized by the diversity of polyphagous species that presented high potential to spread CABMV.ConclusionThe low abundance and diversity of aphid species did not interfere negatively in the CABMV epidemiology. The genus Aphis, particularly Aphis fabae/solanella and A. gossypii, was crucial in the spread of CABMV in passion fruit orchards in the eastern State of São Paulo.
