ABSTRACT.Abies koreana is an endemic and rare species from Korea and is classified as endangered by the International Union for Conservation of Nature. Although the genetic diversity assessment for current population of A. koreana needs to be performed urgently, no microsatellite markers have been developed for this species. In the present study, we developed 22 novel polymorphic microsatellite loci and the characteristics of these loci were determined in A. koreana as well as in Abies nephrolepis, the most closely related species, and these loci were compared with previously reported microsatellite markers developed for the Abies genus. Genomic sequence (161 Mbp; 325,776 reads) was obtained from one individual of A. koreana using Roche 454 GS-FLX Titanium sequencing and 19,258 repeat motifs were identified from it. A total of 288 primer pairs with high copy numbers of direpeat motifs were evaluated for amplification in A. koreana and A. nephrolepis. A total of 71 primer pairs successfully amplified fragments, of which 22 showed polymorphisms in A. koreana and A. nephrolepis. The average expected diversity was 0.767 and 0.717 in A. koreana and A. nephrolepis, respectively; these heterozygosity levels were moderate compared to the previously reported microsatellite loci from Abies species. This is the first set of microsatellite markers developed for A. koreana as well as A. nephrolepis and further population genetic studies of both species and genetic delimitation can be carried out for the species conservation and management.

SummaryThe six Abies species, A. laciocarpa, A. veitchii, A. sachalinensis, A. mariesii, A. faxoniana and A. georgei, were investigated for their chromosomes by the fluorescent banding method using fluorochromes chromomycin A 3 (CMA) and 4′,6-diamidino-2-phenylindole (DAPI). All six species have 2n=24 chromosomes and a similar karyotype consisting of seven pairs of long metacentric chromosomes and five pairs of shorter submetaand subtelocentric chromosomes, supporting previous studies. Eight clear CMA-bands appeared at the interstitial region of one arm of long metacentric chromosomes in all species, and in A. veitchii and A. sachalinensis, their number varied from six to eight among plants and/or populations. A weak CMA-band appeared at the interstitial region of one chromosome pair, while a weak CMA-band appeared at the proximal region in some species. In most species DAPI did not produce clear bands, and sites of clear CMA-bands were DAPI negative. Only A. mariesii showed many DAPI-bands at the interstitial and/or centromeric regions of several chromosomes. A few weak DAPI-bands appeared in some other species. The fluorescent banding and FISH patterns reported in Abies species were compared and discussed with taxonomic treatment and molecular phylogeny of Abies.

Some introduced species cause severe damage, although the majority have little impact. Robust predictions of which species are most likely to cause substantial impacts could focus efforts to mitigate those impacts or prevent certain invasions entirely. Introduced herbivorous insects can reduce crop yield, fundamentally alter natural and managed forest ecosystems, and are unique among invasive species in that they require certain host plants to succeed. Recent studies have demonstrated that understanding the evolutionary history of introduced herbivores and their host plants can provide robust predictions of impact. Specifically, divergence times between hosts in the native and introduced ranges of a nonnative insect can be used to predict the potential impact of the insect should it establish in a novel ecosystem. However, divergence time estimates vary among published phylogenetic datasets, making it crucial to understand if and how the choice of phylogeny affects prediction of impact. Here, we tested the robustness of impact prediction to variation in host phylogeny by using insects that feed on conifers and predicting the likelihood of high impact using four different published phylogenies. Our analyses ranked 62 insects that are not established in North America and 47 North American conifer species according to overall risk and vulnerability, respectively. We found that results were robust to the choice of phylogeny. Although published vascular plant phylogenies continue to be refined, our analysis indicates that those differences are not substantial enough to alter the predictions of invader impact. Our results can assist in focusing biosecurity programs for conifer pests and can be more generally applied to nonnative insects and their potential hosts by prioritizing surveillance for those insects most likely to be damaging invaders.
