Phylogenetic Relationships between Ten genotype of Hordeum vulgare L. Using Molecular Markers

Abstract: and three landraces from Sinai (El-Kheroba, El-Sheikh Zuwaid, and Wadi Sedr) were carried out using two molecular genetic markers (RAPD-PCR and ISSR-PCR). The genetic distance between Ten genotype was also estimated from banding patterns Twenty two random primers were used in RAPD revealed 316 bands while 159 bands were detected of ISSR analysis using 10 primers. RAPD analysis among ten genotypes showed 41.77% polymorphism, while ISSR analysis showed 62.02% polymorphism. It was found that, ISSR was a more viab… Show more

“…RAPD was also found to be an effective tool in detecting genetic diversity in peanut cultivars [18]. El Atroush et al [19] compared RAPD and ISSR's efficacy and found that both RAPD and ISSR markers are suitable tools, but ISSR is a more viable marker than RAPD in the detection of the genetic variability among Hordeum vulgare (barley) varieties. The unbiased heterozygosity (uH e ) ranged from 0.41 with the RMN-P3 primer to 0.16 with the primer OP-B18, with a mean record of 0.23 with ISSR; the uH e ranged from 0.0 with the UBC-810 primer to 0.33 with the UBC-812 and UBC-814 primers, with an average of 0.21.…”

“…They also concluded that the RAPD marker can be a significant tool in in peanut breeding programs. Phylogenetic relationships among ten Hordeum vulgare L. varieties were tested using RAPD-PCR and ISSR-PCR by El Atroush et al [19]. RAPD-PCR showed a 41.77% polymorphism, while ISSR-PCR showed a 62.02% polymorphism.…”
