Photosensitizer‐Incorporated Quadruplex DNA‐Gated Nanovechicles for Light‐Triggered, Targeted Dual Drug Delivery to Cancer Cells

Abstract: A novel light-operated vehicle for targeted intracellular drug delivery is constructed using photosensitizer-incorporated G-quadruplex DNA-capped mesoporous silica nanoparticles. Upon light irradiation, the photosensitizer generates ROS, causing the DNA capping to be cleaved and allowing cargo to be released. Importantly, this platform makes it possible to develop a drug-carrier system for the synergistic combination of chemotherapy and PDT for cancer treatment with spatial/temporal control. Furthermore, the i… Show more

“…The absorbance of formazan (produced by the cleavage of MTT by dehydrogenases in living cells) is directly proportional to the number of live cells 27 . For each MTT assay, 96 culture wells of A549, HeLa, HL7702, and L929 cells were prepared (~5000 cells/well).…”

“…440 The intracellular release of DOX from DNA-capped mesoporous nanoparticles using photogenerated ROS species has also been demonstrated. 441 In order to realize the potential of these stimuli-responsive NPs fully, toxicity must be evaluated in cells and animals.…”

“…28,29 Among the stimuli-responsive controlled release DDSs, light-induced release DDSs have been attracted more and more attention due to their high spatial/ temporal resolution in vivo. [30][31][32] Herein, in this study, a PNL was designed and synthesized. Compared to the reported photo-activated DDSs with complex synthesis process, 33,34 the preparation of the liposome-based PNLs is very simple.…”
