Photodynamic opening of blood-brain barrier

Abstract: Photodynamic treatment (PDT) causes a significant increase in the permeability of the blood-brain barrier (BBB) in healthy mice. Using different doses of laser radiation (635 nm, 10-40 J/cm) and photosensitizer (5-aminolevulinic acid - 5-ALA, 20 and 80 mg/kg, i.v.), we found that the optimal PDT for the reversible opening of the BBB is 15 J/cm and 5-ALA, 20 mg/kg, exhibiting brain tissues recovery 3 days after PDT. Further increases in the laser radiation or 5-ALA doses have no amplifying effect on the BBB per… Show more

“…Note that high radiant exposure (40 J/cm 2 ) caused severe vasogenic edema after PDT for both young and adult mice. This fact suggests that high radiant exposure induces BBB disruption beyond the brain reserve capability, which agrees with our previous results [7].…”

“…In our previous studies on adult mice, we demonstrated that PDT temporally increases BBB permeability . Our results are consistent with data of Hirschberg et al and Madsen et al, who also showed PDT‐related BBB opening using a 400 μm bare flat‐end quartz fiber (635 nm) with 5‐ALA and 670 nm laser application with aluminum phthalocyanine disulfonate (AlPcS2a) .…”

“…Indeed, currently over 70 different methods were suggested for overcoming BBB including physical, chemical and biological approaches [4][5][6] . Nevertheless, these methods are not widely applied in daily clinical practice due to many reasons including invasiveness (e.g., photodynamic opening of BBB that requires skull opening) 7 , challenge in performing (intraarterial injection of mannitol that only few specialists in clinics can usually do) 8 , limitation of drug concentration (intranasal drug delivery) 9 or small area of treatment (only 1-3 mm at usage of focused ultrasound that opens BBB with additional using of micro-bubbles) 10 . All these methods require further studies to improve the reproducibility and technological robustness as well as quantitative evaluation of BBB permeability.…”
