Photodynamic action of some sensitizers by photooxidation of luminol

Abstract: We report the development of a novel simple experimental method which allows the comparison of new photosensitizers based on their production of reactive oxygen species. A high-performance liquid chromatography (HPLC) assay permits the monitoring of several substances (sensitizer, reactant and oxidized end product) simultaneously on a single chromatogram. Photoreactions were monitored throughout their course by the HPLC assay surveying the sensitizers' efficiency of singlet oxygen production by the oxidative d… Show more

“…Rose bengal (4,5,6,7-tetrachloro-2Ј,4Ј,5Ј,7Ј-tetraiodofluorescein) generates ROI in medium when exposed to light (Wierrani et al, 2002). When added to germination medium at 6 M, it resulted in an almost total block in growth of bos1 seedlings ( Figure 4A).…”

“…Paraquat (methyl viologen) promotes the formation of ROS by diverting electrons from photosynthetic electron transport to reduce O 2 to superoxide (Suntres, 2002). Rose bengal (4,5,6,7-tetrachloro-2#,4#, 5#,7#-tetraiodoflorescein) is a photosensitizer that generates singlet oxygen when exposed to light (Wierrani et al, 2002). As shown in Figure 4A, treatments of 10 mM paraquat or 10 mM Rose bengal resulted in the increase of endogenous mRNA levels of AtHEMA2 and AtFC1.…”

“…Reactive Oxygen Species (ROS) Generation Assay. High-performance liquid chromatography (HPLC) assays for the determination of the production of ROS (mainly singlet oxygen species) 11 by FWAs were performed as follows: An Ultimate 3000 system (Dionex Corporation, Sunnyvale, CA) equipped with a 250 × 4.6 mm inner diameter Hi-Pore RP-304 reversed-phase column (Bio-Rad Laboratories, Inc., Hercules, CA), along with a guard column, was used for this assay. An isocratic mobile phase containing acetonitirile and Na 2 HPO 4 buffer (0.033 mol/L, pH 8) in the ratio of 7:15 was used at a flow rate of 0.5 mL/min.…”

“…This HPLC assay is based on monitoring singlet oxygen generation via the degradation of luminol to aminopthalic acid. 11 Oxidative decomposition of luminol was monitored in the presence of the inorganic nanoparticles, rose bengal (a wellknown photosensitizer), 11 and Uvitex NFW, before and after exposure to 1, 6, and 12 h of UV irradiation.…”
