Phosphoproteomic analysis reveals compensatory effects in the piriform cortex of VX nerve agent exposed rats

Abstract: To gain insights into the toxicity induced by the nerve agent VX, an MS-based phosphoproteomic analysis was carried out on the piriform cortex region of brains from VX-treated rats. Using isobaric tag based TMT labeling followed by titanium dioxide enrichment strategy, we identified 9975 unique phosphosites derived from 3287 phosphoproteins. Temporal changes in the phosphorylation status of peptides were observed over a time period of 24 h in rats exposed to a 1× LD50, intravenous (i.v.) dose with the most not… Show more

“…Peptide samples were labeled using TMT 10 plex reagents (Thermo Scientific, Bremen, Germany) as described previously41. Peptides derived from all OKF6/TERT1 were labeled with TMT tag 127N, JHU-O28 with 128C, JHU-O11 with 129N, FaDu with 129C, CAL 27 with 130N, JHU-O22 with 130C and JHU-O29 with 131.…”

“…Multiplexed quantitation via isobaric labeling reagents [e.g., tandem mass tags (TMT) ( 14 ) and isobaric tags for relative and absolute quantitation (iTRAQ) ( 15 )] combined with phosphopeptide enrichment has been employed to dissect complex signaling pathways using mass spectrometry. Nirujogi et al combined an 8-plex TMT labeling strategy with titanium dioxide-based phosphopeptide enrichment to examine the changes of the phosphoproteome in the brains of rats after the exposure to VX, a nerve agent ( 16 ). Roitinger and colleagues combined a 4-plex iTRAQ labeling strategy with a phosphopeptide enrichment pipeline (immobilized metal affinity chromatography coupled with metal oxide affinity chromatography) to characterize the DNA damage response signaling pathway in Arabidopsis thaliana ( 17 ).…”

“…The dissolved peptide was incubated with 0.8 mg of TMT reagent at room temperature for 1 h. The labeling efficiency was determined by pooling 1 μg of amount from each condition and analyzing it on mass spectrometer. The labeled peptides were pooled and fractionated by high‐pH basicRPLC (bRPLC) on an Agilent 1100 LC system as described earlier . A total of 96 fractions were collected and concatenated to 12 fractions and then vacuum‐dried.…”

“…Thus, the identification of strain‐specific proteins using high‐resolution MS‐based approach is an effective strategy for the strain‐level identification and quantification of proteins. We employed isobaric tag based multiplexing strategy for the relative proteomic comparison of the ATCC E. coli Big‐Six strains . We also carried out bioinformatics analysis to identify strain‐level proteins by mapping the MS‐derived data to the genome sequence of the studied E. coli strains.…”
