Phenylalanyl-tRNA synthetase of Escherichia coli K 10. Multiple enzyme.cntdot.aminoacyl-tRNA complexes as a consequence of substrate specificity

Abstract: The interaction between Phe-tRNA(Phe) or other acyl-tRNA derivatives thereof and phenylalanyl-tRNA synthetase of Escherichia coli K 10 has been investigated by nonequilibrium dialysis, by fluorescence titration in the presence of 2-p-toluidinylnaphthalene-6-sulfonate, by the kinetics of the aminoacylation of tRNA(Phe), and by the kinetics of the catalytic hydrolysis of Phe-tRNA(Phe). Phe-tRNA(Phe), or derivatives thereof, forms two types of complexes with the synthetase. One type involves the attachment of the… Show more

“…Kinetics of Enzyme•Acyl-tRNA Complexes. Phenylalanyl-tRNA synthetase complexes of acyl-tRNAs were observed by their ability to decrease the fluorescence intensity of TNS (Güntner & Holler, 1979;Holler, 1980). When TNS occupies the Phe-specific binding site of the enzyme, it becomes highly fluorescent.…”

“…Nonequilibrium Dialysis. The method of Colowick & Womack (1969) has been previously used to measure the stoichiometry of enzyme-aminoacyladenylate complex or the binding of amino acids or aminoacyl-tRNA to aminoacyl-tRNA synthetases (Kosakowski & Bóck, 1971;Hyafil et al, 1976; Güntner & Holler, 1979). Here this technique was applied to the determination of the kinetic rate constant for the dissociation of the phenylalanyladenylate-enzyme complex.…”

“…The adduct of Ap4A with cis-diamminedichloroplatinum(II) has been prepared by the method of Segal et al (1984) and purified similarly to Ap4A (Pimmer et al, 1976). Non-equilibrium dialysis Dissociation constants and stoichiometries were measured by non-equilibrium dialysis as described previously (Colowick & Womack, 1969;Guntner & Holler, 1979;Muise & Holler, 1985). A dialysis apparatus was employed that consisted of a 1500 1dl (upper) reaction…”
