Phenotypic and Functional Characterization of Monoclonal Antibodies with Specificity for Rhesus Macaque CD200, CD200R and Mincle

Abstract: Lectin-like molecules and their receptors are cell surface molecules that have been shown to play a role in either facilitating infection or serving as transporters of HIV/SIV in vivo. The role of these lectin-like molecules in the pathogenesis of HIV/SIV infection continues to be defined. In efforts to gain further insight on the potential role of these lectin-like molecules, our laboratory generated monoclonal antibodies (mAb) against the human analogs of rhesus macaque CD200, CD200R and Mincle, since the rh… Show more

“…Mincle protein is barely detectable on resting cells ( 4 , 42 ). In experiments in various tissues in rhesus macaques, the frequencies of CD14 + gated cells that express Mincle in colon and ileum were low compared with bone marrow, liver, spleen, and lymph nodes ( 43 ). Induction of Mincle expression was shown to be induced by several pathogenic and non-pathogenic stimuli.…”

“…The raw data was then subjected to analysis using the FlowJo Software (ver.9.6.2) (Ashland, OR). The gating strategies utilized for defining the various cell lineages has been published elsewhere (Byrareddy et al , 2014; Byrareddy et al , 2015; Takahashi et al , 2014). …”

“…Mononuclear cells were isolated from the PBMCs and colorectal biopsy specimens as described previously (Byrareddy et al , 2016; Byrareddy et al , 2015). In brief, heparinized blood samples were obtained and mononuclear cells were isolated using ficoll-hypaque gradient centrifugation.…”
