Phenotypic and functional analysis of lymphocytes infiltrating osteolytic tumors: use as a possible therapeutic approach of osteosarcoma

Abstract: Background: Osteosarcoma is the most common type of primary bone tumor. The use of aggressive chemotherapy has drastically improved the prognosis of the patients with nonmetastatic osteosarcomas, however the prognosis of the patients with metastasis is still very poor. Then, new and more effective treatments for curing osteosarcoma, such as immunotherapy are needed. Tumor-infiltrating lymphocytes (TIL) have been involved in the control of tumor development and already assessed with success for the treatment of… Show more

“…However obtained levels were too low in regard to the requirement for immunotherapy [83,84]. In addition, while expanded TILs had higher cytotoxic properties against allogeneic and autologous tumor cells compared to autologous blood peripheral lymphocytes ( Figure 1A), a low expression of immunomodulatory molecules on osteosarcoma cells or/and the secretion of suppressive mediators may prevent activation and expansion of these TILs [78].…”

“…Preclinical data suggest a role for tumor-infiltrating lymphocytes in osteosarcoma (25,26), while several cytokines, including IL-12, IL-2, and type I interferons, have shown preclinical activity (27)(28)(29). Adjuvant use of muramyl tripeptide phosphatidylethanolamine (mifamurtide), a synthetic lipophilic analog of a mycobacterial cell wall protein, was recently shown to improve survival of patients with osteosarcoma (30) and has been approved for clinical use.…”

“…About 20% of patients present visible metastases with imaging at diagnosis and about 25% patients have metastases during the course of treatment. 1 Once the metastases, especially pulmonary metastases, occur it is difficult to treat them due to their poor prognosis. Currently, the characteristics of high metastasis in osteosarcoma cell lines remain unclear.…”
