Phase II trial of idarubicin in patients with advanced lymphoma

Abstract: A phase II trial of idarubicin was performed in 24 patients with advanced lymphoma. The drug was administered in a dose of 10-15 mg/m2 i.v. or 15-70 mg/m2 p.o. (single dose) every 3 weeks. There were four partial responses and four minor responses. All but one of the responders had received prior doxorubicin therapy. The toxicities were myelosuppression, nausea and vomiting, and alopecia. Two patients with compromised cardiac function were observed to have further deterioration in the ejection fraction as meas… Show more

“…Idarubicin has been of special interest for the treatment of high-grade NHL in elderly patients because of the following reasons: (i) When applied in an oral form, the substance has demonstrated efficacy in both chemonaive (response rates 31-86% of cases [115,116]) as well as in pretreated patients (response rates 17-26% [117,118]). …”

“…A depletion of sodium and calcium within the crust with respect to the pristine glass and a more or less marked enrichment for aluminum, iron and manganese is generally reported for decay products of potash-lime and soda-lime glass [4,9,19,20,22,24,28]; less agreement arises for potassium and magnesium, since either depletion [4,9] or enrichment [20,28] were reported for both of them, or even a pronounced enrichment in potassium in the crust, without mention of the magnesium behavior [22]. A thorough work on the decay of archaeological soda-lime medieval glass further suggests that sodium may be more easily leached out of the glass, or less easily incorporated in the decay phases, with respect to potassium [24].…”

“…The drug is approved for the treatment of acute leukemia, and has significant activity in non-Hodgkin's lymphoma. 22,23 In fact, a small phase III trial demonstrated that substitution of idarubicin (10 mg/m 2 ) for doxorubicin (50 mg/m 2 ) in combination with cyclophosphamide, vincristine, and prednisone resulted in an equivalent complete response (CR) rate with less cardiac toxicity in patients with intermediate-grade nonHodgkin's lymphoma. 24 Furthermore, Minderman et al 25 have reported that idarubicin exhibited greater schedule dependence favoring protracted exposure than doxorubicin in a K562 human leukemia cell line.…”
