The Mikania genus has been known to possess numerous pharmacological activities. In the present study, we aimed to evaluate the interaction of 26 selected constituents of Mikania species with (i) cyclooxygenase 2 (COX 2), (ii) human neutrophil elastase (HNE), (iii) lipoxygenase (LOX), matrix metalloproteinase ((iv) MMP 2 and (v) MMP 9), and (vi) microsomal prostaglandin E synthase 2 (mPGES 2) inhibitors using an in silico approach. The 26 selected constituents of Mikania species, namely mikamicranolide, kaurenoic acid, stigmasterol, grandifloric acid, kaurenol, spathulenol, caryophyllene oxide, syringaldehyde, dihydrocoumarin, o-coumaric acid, taraxerol, melilotoside, patuletin, methyl-3,5-di-O-caffeoyl quinate, 3,3′,5-trihydroxy-4′,6,7-trimethoxyflavone, psoralen, curcumene, herniarin, 2,6-dimethoxy quinone, bicyclogermacrene, α-bisabolol, γ-elemene, provincialin, dehydrocostus lactone, mikanin-3-O-sulfate, and nepetin, were assessed based on the docking action with COX 2, HNE, LOX, MMP 2, MMP 9, and mPGES 2 using Discovery Studio (in the case of LOX, the Autodock method was utilized). Moreover, STITCH (Search Tool for Interacting Chemicals), physicochemical, drug-likeness, and ADMET (Absorption, Distribution, Metabolism, Excretion, and Toxicity) analyses were conducted utilizing the STITCH web server, the Mol-inspiration web server, and Discovery Studio, respectively. In the present study, STITCH analysis revealed only six ligands (dihydrocoumarin, patuletin, kaurenol, psoralen, curcumene, and nepetin) that showed interactions with human proteins. Physicochemical analysis showed that seventeen ligands complied well with Lipinski’s rule. ADMET analysis showed eleven ligands to possess hepatotoxic effects. Significantly, the binding free energy estimation displayed that the ligand methyl-3, 5-di-O-caffeoyl quinate revealed the highest binding energy for all the target enzymes, excluding LOX, suggesting that this may have efficacy as a non-steroidal anti-inflammatory drug (NSAID). The current study presents a better understanding of how Mikania is used as a traditional medicinal plant. Specifically, the 26 ligands of the Mikania plant are potential inhibitor against COX 2, HNE, LOX, MMP 2, MMP 9, and mPGES 2 for treatments for acute and/or chronic inflammatory diseases.

Some of the vernacular or scientific names are related to possible medicinal and/or toxic properties that can reveal the presence of potential bioactive agents, contributing to the discovery of new drugs and/or knowledge of the risks associated with their use. This study sought to list the scientific and vernacular names of plants whose lexicons are related to those possible properties of plants and to compare them with the “ethno” (ethnobotanical and ethnopharmacological) and pharmacological data available in the scientific literature. A floating reading of the two classical and reference works on Brazilian medicinal plants was performed, and plants with vernacular and/or scientific names related to the possible medicinal and/or toxic properties were listed. Correlations between the meanings of the species’ names (lexicon) and their possible biological properties were made from their translation from Latin by consulting dictionaries. A bibliographic survey was conducted on the “ethno” and pharmacological data for each species. Finally, data from these three dimensions (lexicon, “ethno,” and pharmacology) were classified and compared using a bioprospection classification. It resulted in a list of 90 plant species belonging to 47 families. 66 of the 90 species presented “ethno” data from the scientific literature, while 46 species presented pharmacological data. Of these, 46 (69.7%) and 27 (58.7%), respectively, showed equivalence with the possible medicinal and/or toxic properties of plants according to their lexicons. According to this study, half of the plants investigated demonstrate equivalence in the three dimensions analyzed (lexicons, “ethno,” and pharmacological data from the scientific literature). Gastrointestinal and nervous system categories are among the most common in all three dimensions. Plant lexicons may be closely linked to the possible medicinal and/or toxic properties and the study of plant lexicons may represent one more approach for the search for new drugs, mainly considering the gastrointestinal, nervous, and parasites categories.

Chemical characterization, antimicrobial activity, and cytotoxic activity of Mikania micrantha Kunth flower essential oil from North East India

Mikania micrantha Kunth (mile-a-minute) is a major invasive weed with medicinal properties. It causes destruction of forests and threat to crop plantations, thus affecting the agriculture-based economy of a country. As an attempt to mitigate this problem, we have studied the applicability of M. micrantha flower essential oil against disease causing microorganisms and inhibition of cancerous cells. Sixty-six compounds were characterized from the flower essential oil by gas chromatography-mass spectrometry. The major constituents were: Isoledene (16%), δ-Cadinene (11.2%), Debromofiliformin (9.4%), Trans-Caryophyllene (9.1%), β-Bisabolene (7.4%), Germacrene-D (5.8%), and Zingiberene (5.3%). The oil is a complex mixture containing mainly sesquiterpene hydrocarbons (62.9%) and oxygenated sesquiterpenes (23.9%) with the other terpene compounds. Well-diffusion and resazurin assay were conducted to determine antimicrobial activity and minimum inhibitory concentration (MIC) of the essential oil. Flower oil showed best activity against Mycobacterium smegmatis and Candida albicans with MIC 8 µg/mL. Moreover, it is the first report of antimicrobial activity of the oil towards M. smegmatis, C. albicans, Staphylococcus aureus, and Pseudomonas aeruginosa. Minimum bactericidal and fungicidal concentration was determined and mechanism of antimicrobial activity was investigated by Field Emission-Scanning Electron Microscopy and Transmission Electron Microscopy analysis. It is the first cytotoxicity report of M. micrantha flower essential oil for cervical, ovarian, and pancreatic cancer cell lines. Through MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay, we have found that oil has excellent cytotoxicity against HeLa cervical cancer cell lines (IC 50 5.44 ± 1.33 µg/mL) and PA1 ovarian cancer cell lines (IC 50 10.57 ± 1.44 µg/mL). The essential oil extracted from the invasive weed M. micrantha flower is now found to have antimicrobial and anticancer properties from our study. Thus, this harmful weed can be utilized as antimicrobial and anticancer agents for the healthcare of human being.
