Cadmium (Cd) is wide-spread toxic metal that pollutes most of the vegetables, which eaten by numerous populations all over the world. The aim of the current work is to evaluate the protective and prophylactic effects of the antioxidant materials in the grape seed extract (GSE) on the hepatic intoxication induced by cadmium chloride toxic material in male Wistar rats. Male adult Wistar rats were divided into four groups. Control group fed on balanced diet and given drinking water. Group two (control positive) given CdCl2 in the dose of 0.44 mg/kg body weight (BW) by stomach tube daily. Group three given grape seed extract GSE in the dose of (100 mg/kg BW) daily by stomach tube. Finally, the fourth group gave mixture of (GSE and CdCl2) by stomach tube in the recommended dose. Blood and liver tissues were collected for further biochemical and histopathological studies. CdCl2 significantly increased the serum levels of malondialdehyde, ALT and AST. Cadmium administration decreased levels of antioxidants (Catalase, GSH-R and GSH-Px). The liver of the control positive groups which given CdCl2 showed degenerative changes in the form of vacuolar and hydropic degeneration. Congestion was evident in the central vein and proliferation of the kupffer cells. These hepatic biochemical and degenerative changes were ameliorated by the co addition of GSE.

Because imperatorin (IPT), the furanocoumarins exhibits antiinflammatory activity, we reasoned that IPT might modulate the allergic rhinitis (AR). The aim of this study was to analyze the regulation of AR by IPT. Here, we show the effect and mechanism of IPT in an ovalbumin (OVA)-induced AR model. The number of rubs after the OVA challenge in the OVA-sensitized mice was significantly higher than that in the OVA-unsensitized mice. The increased number of rubs was inhibited by the oral administration of IPT. The increased levels of IgE and histamine in the OVA-sensitized mice were reduced by IPT administration. The levels of interferon-␥ were enhanced, whereas the levels of interleukin (IL)-4 were reduced on the spleen tissue of the IPT-administered AR mice. Protein levels of IL-1␤, macrophage inflammatory protein-2, intercellular adhesion molecule-1, and cyclooxygenase-2 were reduced by IPT administration in the nasal mucosa of the OVA-sensitized mice. In the IPT-administered mice, the number of eosinophils and mast cells infiltration increased by OVA-sensitization were also decreased. In addition, IPT inhibited caspase-1 activity in the same nasal mucosa tissue. In activated human mast cells, the receptor-interacting protein 2 (RIP2), IB kinase (IKK)-␤, nuclear factor-B (NF-B)/ RelA, and caspase-1 activation were increased, but increased RIP2, IKK-␤, NF-B/RelA, and caspase-1 activation were inhibited by the treatment of IPT. In addition, IPT inhibited caspase-1 activity and IL-1␤ production in IgE-stimulated bone marrowderived mast cells. We can conclude that IPT exerts significant effects by regulating of caspase-1 activation in AR animal and in vitro models.

In our present research, we investigated the hepatoprotective and hepatocurative effects nabk honey in penicillin-induced hepatotoxicity. Biochemical analysis of serum was done for all groups. Hepatotoxicity was confirmed by comparing the serum levels of AST, ALT, ALP, total protein and albumin in penicillintreated group with that of normal salinetreated groups. Nabk honey considerably ameliorated the toxic effects of penicillin on livers. Nabk honey showed the ability to avert the elevated serum AST, ALT and ALP levels, and augmented the total protein and albumin, along with improved histopathological changes in livers. On comparing between hepatoprotective and hepatocurative effects, hepatoprotective effect of nabk honey showed a considerably significant [P < 0.01] improvement in biochemical parameters and morphological changes of livers in penicillin-induced hepatotoxicity. Accordingly, hepatoprotective effect of nabk honey was more effective than hepatocurative effect of nabk honey.
