Hyperlipidemia and oxidative stress are major risk factors for the development of atherosclerosis and cardiovascular disease. This study was designed to investigate the hypolipidemic and hepatic antioxidant potentials of the methanol Leek extract (LE) in a high-fat diet (HFD)-fed rat model. Wistar rats were divided into 6 groups (n = 6/groups) as 1) control fed standard diet, 2) HFD, 3) HFD + (Atorvastatin) (20 mg/kg), and HFD + LE (100, 200, or 300 mg/kg). All treatments were administered by gavage and conducted for 6 weeks, daily. At all tested, but in a dose-independent manner, LE significantly reduced final body weights and food intake and suppressed serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST). Besides, in inhibited serum and hepatic levels of total cholesterol (TC), triglycerides (TGs), and low-density lipoprotein cholesterol (LDL-C) but increased serum and hepatic levels of high-density lipoprotein cholesterol (HDL-C). Concomitantly, LE, at all tested doses increased the activity of superoxidase dismutase (SOD) and glutathione peroxidase (GPx), increased the levels of catalase (CAT) and decreased the levels of malondialdehyde (MDA) in the livers of HFD-fed rats. In conclusion, LE exerts potent hypolipidemic and hepatic antioxidant potentials in HFD-fed rats which highlight its beneficial effects in various metabolic-related disorders.

Long-term consumption of sodium arsenite contaminated water can cause endemic arsenic disease. The proteome profile changes of liver fibrosis after exposure to arsenite containing water remain unclear. In this study, Sprague-Dawley (SD) male rats were treated with sodium arsenite (iAs3+), using a daily dose of 1.36 mg/kg body weight (medium dose group, M), 2.73 mg/kg body weight (high dose group, H) or deionized water (control group, C). Isobaric tags for relative and absolute quantitation (iTRAQ) were used to identify the different abundant proteins (DAPs) after arsenic-induced liver fibrosis. A total of 2987 high-quality proteins were detected (95% confident peptides ≥ 2), 608 of which were differentially expressed (fold change > 2 and p < 0.05) in M group and 475 in H group. Moreover, 431 DAPs were found in both M and H groups and used in subsequent bioinformatic analyses. Gene ontology (GO) analysis revealed 4,709 GO terms could be mapped, among which purine binding, actin filament binding and protein kinase binding were the most enriched terms for molecular function category. In addition, protein-protein interaction analysis showed six clusters of interaction networks. Our data provided new insights into the proteome changes after arsenic-induced liver fibrosis in model rats.

This study investigated the in vitro and in vivo antioxidant activities of polyphenol extracted from black garlic. Black garlic polyphenol (BGP) was extracted from black garlic. The in vitro antioxidant activities of BGP were determined using DPPH·, OH and O 2-radical scavenging assays. The in vivo antioxidant activities were determined by detecting the malondialdehyde (MDA) content and superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities in mice. Results showed that, the DPPH· radical inhibition rate of 200 and 250 μg/mL BGP was equivalent with Vc (P > 0.05). With concentration of 400 and 500 μg/mL, the OH radical inhibition rate of BGP was slightly lower than Vc (P > 0.05). The O 2-radical inhibition rates of 200, 400, 600, 800 and 1000 μg/mL BGP were significantly lower than Vc (P < 0.05). In the groups treated with BGP with suitable dose, the serum MDA content was significantly decreased compared with model group (P < 0.05), and the serum SOD and GSH-Px activities were significantly increased (P < 0.05). BGP has obvious DPPH· and ·OH radical scavenging activities, and can significantly decrease the serum MDA content in mice with oxidative damage, and increase the serum SOD and GSH-Px activities.
