Pharmacokinetics of Theophylline in Children With Asthma

Abstract: The pharmacokinetics of theophylline following intravenous injection of aminophylline, 4 mg/kg of body weight, were determined in 30 children with asthma and in 6 normal adult volunteers. The average total clearance of theophylline was 87 ml/hr/kg in the children and 57 ml/hr/kg in the adults. The biologic half-life of theophylline in the children ranged from 1.42 to 7.85 hours, reflecting mainly pronounced interindividual differences in the elimination rate constant of the drug. There was no significant diffe… Show more

“…The low trough and peak STL in our study probably reflects a lower dosage schedule (14 mg/kg/day) than used by some other workers 13 , 14 . In addition, this could well be due to an interindividual and day‐to‐day variation in absorption and clearance of theophylline 15 . However, the mean variation between the dosage was only 3.06 μ g/ml, showing a fairly even plasma concentration during the 24‐hour period.…”

“…Oral theophylline preparations have a well established role in the management of adult and childhood asthma 1 , 2 , 3 . Because of the short half‐life of plain theophylline, frequent six‐hourly doses are required to maintain therapeutic blood levels 4 . Rapid absorption from the gastrointestinal tract leads to large swings in plasma theophylline levels and, as a result, the initial high peak values are often associated with unacceptable side effects 5 .…”
