Current investigation was to evaluate antidiarrhoeal effect of Casuarina equisetifolia. Anti-diarrhoeal activity of methanol bark extract of Casuarina equisetifolia (CEME) studied on experimentally induced diarrhea in rats, castor oil-induced diarrhoea & enteropooling, meal transit time, and PGE2induced entropooling. The CEME orally administered at doses 100, 200, and 400 mg/kg/atropine 0.1 mg/kg/ received control, respectively, were administered 30 min prior to the administration of castor oil. The CEME 100, 200 and 400 mg/kg/atropine 0.1 mg/kg/ received control to rats fed with as charcoal meal. Thirty minutes later, the intestinal distances moved by the charcoal were measured. The PGE2-induced enteropooling rats were treated with CEME orally at 100, 200, and 400 mg/kg/loperamide 2.5 mg/kg/1 ml of 5% v/v ethanol in normal saline, afterward PGE2 was administered orally to each rat, and intestine fluid volume was measured. The CEME 100, 200 and 400 mg/kg produced inhibition on the frequency of defecation by 33.89%, 42.66%, 59.64 %) and in diarrhoeic drops 35.83%, 43.06%, 49.72%. In PGE2-induced entropooling, intraluminal fluid accumulation reduced significantly. The charcoal meal transit time was significantly reduced, at CEME 100 mg/kg. Further at CEME 400 mg increased intestinal motility charcoal meal. The result shows that administration of CEME in rats causes antidiarrhoeal effect by reducing the total number of faeces, water content of faeces,inhibition of defecation, inhibition of diarrhoeic drops, and weight of intestinal content intestinal fluid volume and also increases the diarrhoeal onset time. From the above observation, it was concluded that CEME extracts exhibit significant dose-dependent antidiarrhoeal activity. INTRODUCTION:Diarrhoea is associated with an increased frequency of bowel movements with the production of soft or watery stools.

Aim: Evaluation of Antioxidant and Anti-Parkinson activity of Portulaca oleracea seed methanolic extract. Place: C. U. Shah College of Pharmacy and Research, Wadhwan, Surendranagar, Gujarat, India. Methodology: Collect plant materials were extracted with methanol. Extract was subjected to qualitative and quantitative investigation and antioxidant properties of extract was determine by Nitric oxide free radical scavenging activity and Reducing power by FeCl3 method. Anti-Parkinson activity evaluated by two behavioral models namely, haloperidol induced catalepsy, and orofacial dyskinesia both models various behavioral activity/ parameter (catalepsy, vacuous chewing movement and tongue protrusion) were evaluated. Results: Preliminary qualitative phytochemical screening was to reveal presence of polyphenols, flavanoids, glycoside, alkaloids, carbohydrates and reducing sugar etc. Based preliminary qualitative phytochemical screening; quantitative estimation of methanolic extract showed significant amount of polyphenols. In-vitro antioxidants was performed by two method reducing power by FeCl3 and nitric oxide free radical scavenging, the methanolic extract shows significant antioxidant properties, based on polyphenols and antioxidant properties extracts was used for the Anti-Parkinson activity Haloperidol induced catalepsy in mice Treatment with Portulaca oleracea seed showed a significant (P<0.01) reduction in the duration of cataleptic behavior dose dependently when compared to haloperidol treated group. Haloperidol induced orofacial dyskinesia in rat recovery of orofacial dyskinesia as evidenced by decrease in the frequency of vacuous chewing movement and tongue significant (P<0.05) decrease in the frequency of vacuous chewing & tongue protrusion while Portulaca oleracea seed (200 mg/kg) was found to be insignificant in this respect. Conclusion: After Portulaca oleracea seed (MLPO) treatment, the significant alterations produced in Parkinson’s affected rodents in respect to lipid peroxidation and antioxidant concentration significantly contributing its antioxidant potential. This antiperoxide action observed in Portulaca oleracea seed (MLPO) treated animals might be due to the suppression of the production of reactive oxygen species. This compound may be found to scavenge free radicals, including hydroxyl anions and reduce the level of lipid peroxidation in MLPO animals. Inhibition of oxidative stress may be one of the possible mechanisms for the anti-Parkinson effects of Portulaca oleracea seed (MLPO).

The polyherbal combination was prepared by using aqueous and methanol root extracts of A. officinalis, B. diffusa, C. papaya, C. fistula, C. intybus, F. hispida, F. indica, C. nurvala, S. virgaurea and V. negundo in equal proportion for the dose of 25 mg/kg and 50 mg/kg body weight. Albino Wistar rats were selected for the study; the serum uric acid, urea, creatinine, BUN, and albumin level were measured using standard procedures. The elevated parameters in serum shows the statistically significant (** p<0.05) improvement with a low dose of aqueous extract combination i.e., hemoglobin (11.15 g/dl), CBC (5.93 ×10 3 /µl), total bilirubin (0.61 mg/dl), total protein (09.85 mg/dl), AST (63.41 IU/L) & ALT (34.37 IU/L), uric acid (1.68 mg/dl), urea (57.06 mg/dl), creatinine (1.08 mg/dl), BUN (41.50 mg/dl) and albumin (40.84 mg/dl) in serum and urine. It was also reported as 24h urinary protein (2.96 mg/24h), albumin (09.23 mg/dl), urea (2.80 mg/dl), creatinine (38.28 mg/dl) and uric acid (126.08 mg/dl) shows significant improvement against disease control at the end of the study. Histological changes show that the gentamicin significant damaged tubular necrosis, epithelial cells, glomerular damage, and congestions of the nephron, which was completely reversed with aqueous extract polyherbal combination. Finally, it was concluded that the low dose (25 mg/kg) of aqueous extract has immense potential as compare to methanol extract and possess protective with therapeutic properties in the treatment of glomerulonephritis.
