Pharmacogenetic Loci for Rosuvastatin are Associated with Intima-Media Thickness Change and Coronary Artery Disease Risk

Abstract: Aim: Polymorphisms at LPA, LDLR, APOE, APOC1, MYLIP and ABCG2 are attractive targets for assessment of their impact on lipid-lowering therapy with rosuvastatin. The present study investigated whether polymorphisms at these genes are associated with the risk of coronary artery disease (CAD) development, and reduction of atherogenic lipids and carotid intima-media thickness (CIMT) in CAD patients, taking rosuvastatin. Materials & methods: 190 CAD patients and 1697 subjects were enrolled in pharmacogenetic an… Show more

“…Despite HF is an irreversible progressive disease, the abnormalities in blood lipids (BL), BP, and BG can be identified early and treated effectively with medications. Previous studies have reported that BL, BP, and BG have potential shared genomic loci with HF (7,8). In addition, the present genetics association study of BL, BP, and BG exhibits a substantial sample size, demonstrating significant polygenic heritability and greater multitude of genomic loci in contrast to prior research.…”

“…TC levels higher than 4.0 mmol/L and LDL-C levels higher than 1.8 mmol/L were considered criteria for dyslipidemia. Biochemical and ultrasound investigations were described in our previous paper [31]. Plasma lipid levels, except LDL-C, were detected using an automatic laboratory analyzer.…”

“…DNA samples were obtained from the biobank of Research Institute for Genetic and Molecular Epidemiology of Kursk State Medical University. The samples were collected previously in the frames of genetic studies of cardiovascular and other chronic diseases for the period from 2003 to 2018 [ 17 , 18 , 19 , 20 , 21 , 22 , 23 ]. A total number of participants comprises 1700 unrelated individuals of Slavic origin, inhabitants of the Kursk region, and included 991 patients with a diagnosis of coronary artery disease (633 men and 358 women, medium age 59.9 ± 8.8 years) who were hospitalized in Cardiology Divisions of Kursk Emergency Hospital and Vascular Surgery Division of Regional Clinical Hospital (Kursk).…”
