Phage-displayed peptides that mimic epitopes of hepatitis E virus capsid

Abstract: Hepatitis E is an emerging zoonotic infection of increasing public health threat for the UK, especially for immunosuppressed individuals. A human recombinant vaccine has been licensed only in China and is not clear whether it protects against hepatitis E virus (HEV) genotype 3, the most prevalent in Europe. The aim of this study was to use phage display technology as a tool to identify peptides that mimic epitopes of HEV capsid (mimotopes). We identified putative linear and conformational mimotopes using sera … Show more

“…Phage particles can also be engineered to carry vaccine antigens on their surfaces or they can be used as a vehicle to deliver DNA vaccines (Clark and March 2004). Such structural vaccines can be used to mimic Hepatitis E viral infections (Larralde and Petrik 2017).…”

“…And the long peptide epitopes are mostly in the form of spatial conformation. Mimic epitope is a structure in which the sequence is from the discontinuous amino acid of a spatial conformation, and it may have homology to the certain antigens (Larralde and Petrik 2017), or may have no similarity to the antigens at all. Screening of mimic epitopes not only avoids the cumbersome synthesis of long peptides and the purification of recombinant proteins, but also overcomes the low affinity of the long chain amino acids to antibodies due to the helix or folding.…”

“…93,94 It is crucial to use vaccine to prevent HEV infection, thus knowledge for the structure of the HEV genome and the function of ORFs are essential for vaccine development. [95][96][97][98] Although four genotypes of HEV have been reported globally, they belong to one serotype. The antibodies induced by different genotype HEV can neutralize homologous or heterologous HEV.…”
