PGC‐1α, a potential therapeutic target against kidney aging

Abstract: Aging is defined as changes in an organism over time. The proportion of the aged population is markedly increasing worldwide. The kidney, as an essential organ with a high energy requirement, is one of the most susceptible organs to aging. It is involved in glucose metabolism via gluconeogenesis, glucose filtration and reabsorption, and glucose utilization. Proximal tubular epithelial cells (PTECs) depend on lipid metabolism to meet the high demand for ATP. Recent studies have shown that aging‐related kidney d… Show more

“…Proliferation of VSMCs caused by vascular injury plays an important role in the formation of vascular lesions (Durham et al, 2018;Zhang C. et al, 2018). Peroxisome proliferatoractivated receptor g coactivator-1 (PGC-1) is a very important regulator of many biological processes such as energy metabolism (Islam et al, 2018;Lee et al, 2019). PGC-1b expression was decreased in the carotid artery of rats with balloon injury (Guo et al, 2013).…”

“…Mitochondrial biogenesis is mainly regulated by a set of nuclear encoded coactivators and transcription factors. Peroxisome proliferator–activated receptor γ coactivator 1α (PGC‐1α), which is critical for maintaining energy homeostasis, has been shown to play a key role in regulating mitochondrial biogenesis and function 41 . Decreased PGC‐1α expression and activity have been linked to cardiac ageing and the development of heart failure 42 .…”

“…The transcriptional coactivator PGC-1α is a deacetylated substrate for SIRT1 and plays a critical role in mitochondrial biosynthesis and maintenance [ 49 , 50 ]. The PGC-1α protein, which is abundant in mitochondria-rich tissues, consists of 795 amino acids with a molecular weight of approximately 92 kDa [ 51 ].…”
