Persistent Infection of a Rat Kidney Cell Line with Rauscher Murine Leukemia Virus

Abstract: The propagation of a murine leukemia virus (Rauscher) in a kidney cell line, derived from a rat with lymphoid leukemia, was studied. A complement-fixing (CF) antigen reacting with Rauscher immune sera was detected at various passage levels, which correlated with the visualization by use of electron microscopy of viral buds and viral particles in different stages of maturation in all passages. Five-month-old monolayers continued to shed virus and to yield high CF antigen titers. The cell-free supernatant fluid … Show more

“…To test for transmissibility of the endogenous viruses, supernatant fluids were filtered through 0.45 p Millipore filters (Millipore Corp., Bedford, Massachusetts, USA) and used to infect the BALB/3T3, NTH/3T3 (Jainchill et a/., 1969), R4, a radiation-transformed derivative of BALB/3T3 (Pollock et al, 1970) and the rat NRK cell lines (Duc-Nguyen et al, 1966).…”

“…All cell lines were grown in Dulbecco's modification of Eagle's minimal essential medium supplemented with 10% fetal calf serum (Gibco, Belgium). The following cell tines were used: the continuous mouse cell lines NIW3T3 and BALB/c 3T3 (Jainchill et al, 1969); NRK, normal rat kidney cells (Duc-Nguyen et al, 1966); Mink Mv 1 Lu cell line (Shannon, 1972); REF, cell line established from an individual embryo of Wistar rat; canine sarcoma D17 (Riggs et al, 1974); F115, Syrian hamster cell line, kindly given by Dr. Aaronson (National Cancer Institute, Bethesda, USA); 65770B/ A6 and 23501/A6, respectively, FBJ-MuSV transformed rat and hamster non-producing cell lines, kindly provided by Dr. J.A. Levy (Levy et al, 1978).…”
