Peptides from Mucuna pruriens L., with protection and antioxidant in vitro effect on HeLa cell line

Abstract: BACKGROUND Mucuna pruriens L. is a legume sown in the Mexican southeast with an important protein content. Studies have shown the potential use of by‐products derived from Mucuna as a functional food because of the hypoglycemic and antihypertensive activities. Thus, this study aims to assess the antioxidant and protective effect of the peptide fractions derived from M. pruriens L., in vitro on the HeLa cell line. An enzymatic hydrolysis with pepsin–pancreatin was performed on the total protein concentrate, fro… Show more

“…Based on the methodology reported by Segura‐Campos, Salazar, Chel, & Betancur (2013), a protein‐rich fraction was obtained, which was subjected to an enzymatic hydrolysis process using a sequential pepsin‐pancreatin system, based on the method reported by Martínez‐Leo, Martín, Acevedo, Moo, and Segura‐Campos (2019). Protein‐rich fraction was adjusted to pH 2 using 6 N HCl and hydrolyzed by treatment with Pepsin (Sigma ® , P7000‐100G) for 45 min, followed by treatment with Pancreatin (Sigma ® , P3292‐100G) for another 45 min after adjusting to pH 7.5 using 6 N NaOH.…”

“…The enzymatic hydrolysis was stopped by increasing the temperature to 80°C for 20 min. In order to eliminate the insoluble portion, the digested sample was centrifuged at 1,317 g for 20 min (Martínez‐Leo et al., 2019).…”

“…The imbalance between oxidizing species and endogenous antioxidants causes oxidative stress. Moreover, oxidative stress has been associated with degenerative diseases like cardiovascular disorders and type II diabetes (Martinez-Leo et al, 2019). The intake of antioxidants could reduce the morbidity and mortality associated with coronary heart diseases, as demonstrated in several studies (Durak et al, 2013).…”
