Peptide Regulation of Skin Fibroblast Functions during Their Aging In Vitro

Abstract: The effect peptides KE, KED, AED and AEDG on proliferation (Ki-67), regeneration and aging (CD98hc), apoptosis (caspase-3), and extracellular matrix remodeling (MMP-9) in skin fibroblasts during their aging in culture were studied by immunofluorescent confocal microscopy. All studied peptides inhibited MMP-9 synthesis that increases during aging of skin fibroblasts and enhanced the expression of Ki-67 and CD98hc that are less intensively synthesized during cell aging. Peptides AED and AEDG suppressed caspase-d… Show more

“…MTT test showed that AED, KED, KE, and AEDG peptides and their combination stimulate hPDLSCs proliferation. It has been reported that KE peptide and AED peptide, which stimulate proliferation of immune cells and fibroblasts, 27,31 can potentiate proliferation activity and have a proliferation effect in hPDLSCs. KED peptide in previous investigation decreased stem cell proliferation, but stimulates thymocytes and skin fibroblast proliferation.…”

“…These short “cryptic” peptides can be formed endogenously by proteolytic cleavage of nuclear proteins or by synthesis after which they penetrate the cytoplasmic and nuclear membrane [60, 61]. These peptides are involved in the epigenetic regulation of aging and can have health-promoting effects by for example suppressing age-related increased expression of matrix metalloproteases and caspase-dependent apoptosis [55, 93].…”
