Peptide–protein interactions studied by surface plasmon and nuclear magnetic resonances

Abstract: The structural features of the complexes that K K-bungarotoxin forms with three different synthetic peptides, mimotopes of the nicotinic acetylcholine receptor binding site, have been compared to the corresponding nuclear magnetic resonance (NMR) and surface plasmon resonance (SPR) data. For the considered peptides, the observed different affinities towards the toxin could not be accounted simply by static structural considerations. A combined analysis of the SPR-and NMR-derived dynamic parameters shows new co… Show more

“…The binding affinities of the full-length and N-terminal deleted SARS 3CL pro s to the substrate peptide were determined using the SPR biosensor technology (21). The measurements were performed using the dual flow cell Biacore 3000 instrument (Biacore AB, Uppsala, Sweden).…”

“…In that respect, values derived from SPR measurements have been shown to be in excellent agreement with those obtained by other biophysical approaches. 10,34 SPR-based affinity measurements have been acknowledged to be extremely helpful in the field of structural biology as one can judge by the increasing numbers of scientific reports in which SPR measurements are correlated to NMR 24, 34,43,55,62 or crystallographic data. 28,31,39,45,61 More specifically, a comparison of the affinities related to the interactions of a protein with its natural partner and with various truncated forms 30,53 or single-point mutants of the latter, is a convenient way to validate conclusions drawn from structural data.…”

“…6 In such cases, as we have suggested recently, a comparison of NMR and surface plasmon resonance (SPR) data of kinetic parameters, such as k on and k off , can be very helpful to correlate the intermolecular interaction with the corresponding structural information in solution. 7 Among the various peptide -protein interactions, the complex formation between acetylcholine receptor (AChR) mimotopes and a-bungarotoxin (a-btx) has been analysed thoroughly in terms of biological activity, kinetics and solution structure. 8 -11 Therefore, it represents a suitable model system for discussing general aspects of several intermolecular association processes.…”

“…1,7,15 Since clear and comprehensive correlations between structural and functional aspects of complexes between AChR mimotopes and a-btx have not been found, the present NMR structure is discussed also in the light of all known a-btx complexes.…”
