This study aims to determine the combination of the hatching egg yolk (HEY) and amniotic fluid (AF) in Tris diluent to the type of goat spermatozoa abnormalities after frozen at temperature -80°C. This study used four treatments and 6 replications. The semen is diluted using tris extender and added treatment, there are control group (T0) (Tris egg yolk 20%), T1 (15% HEY + 5% AF), T2 (10% HEY + 10% AF), and T3 (5% HEY + 15% AF). After dilution, the semen was equilibrated for 2 hours, then stored using Mr. Frosty and frozen at temperature -80°C in the ultra-freezer for 24 hours. The type of spermatozoa abnormalities type was evaluated on fresh semen and after thawing. The data were analyzed using ANOVA (P<0.05). The results showed that there is no significant difference among treatments. The primary abnormalities in fresh semen, there were macro-cephalic, amorphous, and the secondary spermatozoa abnormalities such as detached head, midpiece defect, coiled tail, loosehead, shoe-hook tail, and a broken tail. The secondary abnormalities in post-thawing were found both in the control group and treatment, which are detached head, loosehead, coiled tail, shoe-hook tail, and broken tail. The conclusion of this study was the percentage of secondary abnormalities after thawing was increasing both in the control and treatment groups.

The purpose of this study was to examine the effect of adding various concentrations of dragon fruit peel extract (DFPE) in Tris-egg yolk diluent (TEY) on the quality of bligon goat spermatozoa. Semen was collected twice a week using the artificial vaginal method from three year old bligon male goats with normal body conditions and reproductive organs. After the macroscopic and microscopic evaluation, good quality semen with a sperm motility of > 70%, a concentration of > 800 x 106 mL-1, and abnormality of < 15 %, was diluted with TEY + DFPE at a concentration of 0% (P0), 2% (P1), 4% (P2), 6% (P3), 8% (P4), dan 10% (P5). The semen which still had a motility ≥ 70% was then stored at a temperature of 3-5oC. Evaluation of the quality of spermatozoa was carried out every 24 hours until the motility decrease to 40%. The results showed that from day 1 to day 5 of preservation, spermatozoa preserved in TEY + 6% DFPE diluent (P3) had higher motility and viability (P<0,05) than other treatments. The abnormalities of spermatozoa until the 5 day of preservation were below 6%, but showed a significant difference (P<0,05) between treatments that received DFPE suplemention with control (P0). It was concluded that the addition of DFPE in TEY up to 6% could reduce the decrease in sperm quality of bligon goats during in vitro preservation.

ABSTRACT Moringa leaves contain various nutrients that can protect spermatozoa during semen preservation. The purpose of this study was to examine the ability of moringa leaf extract with different concentrations of coconut water extender on the quality of Boer crossbreed goat spermatozoa preserved at 3-5°C. The treatment of this study was the concentration level of moringa leaf extract in coconut water extender. Fresh semen was divided into five tubes and diluted with 80% coconut water + 20% egg yolk (P0, control), 79% coconut water + 20% egg yolk + 1% moringa leaf extract (P1), 77% coconut water + 20% egg yolk + 3% moringa leaves ectract (P2), 75% coconut water + 20% egg yolk + 5% moringa leaf extract (P3), and 73% coconut water + 20% egg yolk + 7% moringa leaf extract (P4), respectively. The variables observed were motility and spermatozoa viability. Results of this study showed that supplementation of different concentrations of moringa leaf extracts in coconut water extender was significantly (P<0.05) on motility and viability of Boer crossbreed goat spermatozoa during preservation. Supplementation of 3% moringa leaf extract in coconut water extender was better than 1, 5, and 7% in maintain the quality of Boer crossbreed goat spermatozoa during preservation.
