Peculiarities of the Effects of Bile Acids on Atpase Activity of the Colon Mucosa in Patients With Overweight and Irritable Bowel Syndrome

Abstract: The aim is to investigate the effect of bile acids on the ATPase activity of the colon mucosa in patients with overweight and irritable bowel syndrome (IBS). Materials and methods: Completely examined 12 patients with IBS and overweight. We estimated the ATPase activity of colon mucous of the patients with IBSspectrophotometrically by determined the content of orthophosphate that was released after ATP hydrolysis. We studied the effect of 3-sulphate of taurolitocholate (TLC-S) on specific activities of Na+/ K+-… Show more

“…This Ca 2+ -sensitive is used to monitor the stored Ca 2+ concentration [2] in the lumen of organelle. Isolation of subcellular postmitochondrial fraction of the rat liver was made as described previously [1,3,5]. Assay of ATPase activity was carried out basing on determining the content of inorganic phosphorus by the spectrophotometric method.…”

“…ATPase activity of these patients’ samples of colon mucus was estimated in post-mitochondrial subcellular fraction which was obtained by separate centrifugations as it was described previously for NK/Ly cells [ 18 ]. Briefly, tissue samples homogenized with a glass–glass homogenizer at 300 rpm for 10 min at 0 °C to 2 °C.…”

Briefly, tissue samples homogenized with a glass-glass homogenizer at 300 rpm for 10 min at 0 • C to 2 • C. The homogenate was centrifuged for 10 min at 3000 g using JouanMR 1812 centrifuge (Jouan, France) to precipitate nuclei, large cells fragments, and undestroyed cells, whereas mitochondria remained in the supernatant 1.…”

“…Isolation of the subcellular post-mitochondrial fraction was performed as previously described for rat liver [23,48,62] and for human colon mucosa and human colorectal samples [17], the determination of ATPase activity was based on the determination of the content of inorganic phosphorus by the spectrophotometric method. ATPase activity was estimated in the post-mitochondrial subcellular fraction obtained by differential centrifugation.…”
