Pectic oligosaccharide structure-function relationships: Prebiotics, inhibitors of Escherichia coli O157:H7 adhesion and reduction of Shiga toxin cytotoxicity in HT29 cells

Abstract: Shiga toxin (Stx)-producing, food-contaminating Escherichia coli (STEC) is a major health concern. Plant-derived pectin and pectic-oligosaccharides (POS) have been considered as prebiotics and for the protection of humans from Stx. Of five structurally different citrus pectic samples, POS1, POS2 and modified citrus pectin 1 (MCP1) were bifidogenic with similar fermentabilities in human faecal cultures and arabinose-rich POS2 had the greatest prebiotic potential. Pectic oligosaccharides also enhanced lactobacil… Show more

“…In addition, GOS significantly reduced the adhesion of the murine bacterial pathogen Citrobacter rodentium to the epithelial cell surface in a dose-dependent manner in vitro, but not in vivo, which might be related to the expression of adhesins that are insensitive to GOS when C. rodentium grows in vivo (Kittana et al, 2018). of Shiga toxin-producing E. coli to human IECs (Di et al, 2017). Moreover, POS concentration dependently inhibited the adherence and invasion of C. jejuni to Caco-2 cells (Ganan et al, 2010).…”

“…POS have the ability to compete with Shiga toxin 2 binding to the neutral glycolipid globotriaosylceramide Gb3 receptor on the human IEC surface. Low MW and de‐esterification of POS facilitates the anti‐adhesive activity of Shiga toxin‐producing E. coli to human IECs (Di et al, ). Moreover, POS concentration dependently inhibited the adherence and invasion of C. jejuni to Caco‐2 cells (Ganan et al, ).…”

“…This may make these NDCs suitable for the supplementation in infant formula in the near future. Recently Di et al (Di et al 2017) studied the potential prebiotic effects of 5 different pectin oligosaccharides (POS) structures, which can be obtained from pectin by enzymatic treatment and acid hydrolysis (Di et al 2017). It was found that POS exhibited prebiotic activity in in vitro batch fermentation studies and enhancing effects on numbers of Bifidobacteria over time.…”

“…However, the bifidogenic effect was not the same for all POSs tested and seems to be dependent on structure. Especially the arabinose-rich rhamnogalacturonic acids were responsible for the prebiotic effects (Di et al 2017 …”

“…For instance, different POS from orange peel have enhanced lactobacilli growth, while POS from orange peel with low MW and low deesterification showed the most anti‐adhesive activity for E . coli O157:H7 (Di et al, ).…”
