Pdx1 and Ngn3 Overexpression Enhances Pancreatic Differentiation of Mouse ES Cell-Derived Endoderm Population

Abstract: In order to define the molecular mechanisms regulating the specification and differentiation of pancreatic β-islet cells, we investigated the effect of upregulating Pdx1 and Ngn3 during the differentiation of the β-islet-like cells from murine embryonic stem (ES) cell-derived activin induced-endoderm. Induced overexpression of Pdx1 resulted in a significant upregulation of insulin (Ins1 and Ins2), and other pancreas-related genes. To enhance the developmental progression from the pancreatic bud to the formatio… Show more

“…Furthermore, another study used human adipose tissue-derived mesenchymal stem cells, initially expressing c-Kit, to induce pancreatic endocrine cell differentiation under stringent in vitro conditions [92]. In mice, PDX-1 and NGN3 overexpression induced formation of immature insulin/C-peptide-producing cells from mouse embryonic stem cell-derived c-Kit + endoderm populations [96]. In addition, c-Kit + cells isolated from the salivary gland of adult mice were differentiated into pancreatic endocrine lineages under glucagon-like peptide 1 treatment [97,98].…”

“…This study suggested that Nkx2.2 promotes differentiation of ESCs into IPCs (Shiroi et al 2005). Kubo et al (2011) reported that the overexpression of Pdx-1 and Ngn3 enhanced pancreatic differentiation. Pdx-1 is involved in early pancreatic development.…”

“…One example application might be to implement an automatic multistep stem cell differentiation protocol without laborious sequential application of chemical inducers (Schiesser et al 2014). For example, a gene circuit could recapitulate the ontogeny of pancreatic b cells by sequentially expressing Gata6, Pdx1, Ngn3, and Pax4 to drive differentiation of stem cells through definitive endoderm (Wamaitha et al 2015), pancreatic endoderm (Kubo et al 2011), and b cells, respectively ( Fig. 1, left) (Collombat et al 2009).…”
