PAX5 alteration-associated gene-expression signatures in B-cell acute lymphoblastic leukemia

Abstract: The paired box domain gene 5 (PAX5) is frequently altered in both childhood and adult B-cell acute lymphoblastic leukemia, and takes part in leukemogenesis. We analyzed data from the database of Gene Expression Omnibus (accession number: GSE11877) using bioinformatical and statistical methods. The results showed that cases of PAX5 alteration can cluster using unsupervised clustering algorithms, and one gene, zinc and ring finger 1 (ZNRF1), was characterized and validated by quantitative RT-PCR. ZNRF1 may be as… Show more

“…But the role of ZNRF1 in biology of cancer has been seldom reported before. Previous research showed that ZNRF1 was associated with leukemia development and downexpression of ZNRF1 might be involved in PAX5 mutation-inducing leukemia (Shang et al 2013). So, ZNRF1 may take part in the regulation of cell proliferation and apoptosis during the development of human cancers.…”

“…CBX8 triggers the progression of hepatocellular carcinoma by increasing the expression of hsa-miR-365a-3p down-regulated ZNRF1 (34). Deletion of ZNRF1 may be involved in the mechanism of B-cell acute lymphoblastic leukemia associated with PAX5 alteration (35). This indicates that ZNRF1 is a tumor suppressor gene.…”

“…Familiades (2009) 119 yetişkin Pro B-ALL hastasında yaptığı çalışmada, hastaların %30'un da PAX5 mutasyonu tespit edilmiştir (63). Shang (2013) pediatrik ve yetişkin B-ALL hastalarında, PAX-5 gen değişimleri taşıyan bireylerde, ZNRF1 ifadelenmesinin önemli ölçüde azaldığını rapor etmişlerdir ve PAX5 değişimleri taşıyan hastalarda ZNRF1'in lösemi gelişimine neden olabileceği görüşünü bildirmişlerdir (64). Shah (2013) öncü B-ALL örneklerinde 2 farklı aileyi oluşturan bireylerde, eksom sekanslama tekniği ile PAX5 geninde tekrarlayan germline bir mutasyon (p.Gly183Ser) tanımlamışlardır, bu mutasyon genin octapeptid bölgesinde olup, bu mutasyonun öncü B-ALL patogenezinde önemli olduğu rapor etmişlerdir.…”

“…Bhücrelerinden plazma hücresi oluşum aşamasına kadar görev ifadelenmesi devam eder. Bu nedenle PAX5'in B hücre gen ifadelenmesinde anahtar gen olduğu düşünülmektedir(56). PAX5, 9. kromozomun kısa kolu 9.p13 te lokalize olmuştur, 10 ekzondan oluşmaktadır (13, 57).…”
