Partially resistant Cucurbita pepo showed late onset of the Zucchini yellow mosaic virus infection due to rapid activation of defense mechanisms as compared to susceptible cultivar

Abstract: Zucchini yellow mosaic virus (ZYMV) is an emerging viral pathogen in cucurbit-growing areas wordwide. Infection causes significant yield losses in several species of the family Cucurbitaceae. To identify proteins potentially involved with resistance toward infection by the severe ZYMV-H isolate, two Cucurbita pepo cultivars (Zelena susceptible and Jaguar partially resistant) were analyzed using a two-dimensional gel electrophoresis-based proteomic approach. Initial symptoms on leaves (clearing veins) developed… Show more

“…Indeed, the up-regulation of genes involved in energy production and cell repairing was observed at 12 DPI. Our results were in agreement with Nováková et al [ 31 ], in which the partially resistant C.pepo cv Jaguar showed a strong up-regulation of several proteins involved in photosystems activities 15 days after ZYMV inoculation [ 31 ]. Interestingly, we found a high up-regulated gene coding for an HSP90.5 (Cup001195g045603.1) with a LogFC of 4.6 exclusively expressed in 381e (GO:0045037, supplementary material 4 ) .…”

“…A). In consequence, our results suggest that, at a very early PPV infection stage (when only a few viral particles are present), infection triggers ROS production, confirming the previous reports on ROS release during viral infections in plants (Allan et al ., ; Díaz‐Vivancos et al ., ; Love et al ., ; Manacorda et al ., ; Nováková et al ., ). However, at a late PPV infection stage (with full‐blown virus accumulation), PPV impairs both the PPV‐ and PTI‐related oxidative burst.…”

“…The alleviating effect of oxidative stress by Hx has been demonstrated in tomato against B. cinerea and P. syringae ( Finiti et al, 2014 ; Camañes et al, 2015 ). Nováková et al (2015) have also reported that resistance of Cucurbita pepo against Zucchini yellow mosaic virus infection is related to a faster accumulation of the proteins involved in ROS detoxification. Our results agree with recent data that support the notion that maintaining a basal level of ROS is essential for normal life ( Mittler, 2017 ).…”
