Palmitoyl ascorbate: Selective augmentation of procollagen mRNA expression compared with L‐ascorbate in human intestinal smooth muscle cells

Abstract: The effect of 6-O-palmitoyl ascorbate on procollagen mRNA levels, collagen synthesis, and collagen secretion was investigated and compared with the effect of L-ascorbate in human intestinal smooth muscle (HISM) cells in vitro. Collagen synthesis, determined by the incorporation of 3H-proline into pepsin-resistant, salt-precipitated collagen, increased in a concentration-dependent manner in response to palmitoyl ascorbate. There was a twofold increase in collagen synthesis at 2.5 and 5 microM. By contrast, L-as… Show more

“…In other cultured human fibroblast experiments, MAP was found to be equivalent to AA in stimulating collagen synthesis, while SAP required at least a tenfold greater concentration to produce the same effect as AA . AA‐Pal augmented human intestinal smooth muscle cells procollagen synthesis in vitro more effectively than AA but induced similar increases in the amounts of newly synthesized collagen secreted into the medium …”

“…58 AA-Pal augmented human intestinal smooth muscle cells procollagen synthesis in vitro more effectively than AA but induced similar increases in the amounts of newly synthesized collagen secreted into the medium. 59 AA also plays a role in collagen synthesis at the level of gene expression. 60,61 It has been shown to up-regulate collagen synthesis and increase the synthesis of the inhibitor of metalloproteinase-I 62 which decreases UV-induced collagen degradation.…”

“…The largest number of matched hits was for ascorbate and aldarate metabolism, phenylalanine, linoleic, and arginine, and proline metabolism. Notable contributions came from metabolites attributed to ascorbate and proline metabolism, suggesting variation in collagen production and thus differences in cell matrix features. Although ascorbate is instrumental in collagen production, it is expected that the variance observed with ascorbate metabolism has been somewhat biased by the inclusion of ascorbate-2-phosphate as a component of the OIM culture media. Linoleic acid metabolism, which contributes to the differences noted between sample types, is known to play an important role in bone development as well as acting as precursor for a class of broad range cell membrane signaling molecules (eicosanoids), inferring significant differences in cellular regulation …”

“…The 16‐carbon chain of PA affects both physico‐chemical and biophysiological properties of this molecule. Thus, PA has greater potency to affect the redox state in mammalian cells [28], alter DNA synthesis in tumor cells and their proliferation [29], suppress the posttranscriptional induction of ornithine decarboxylase [30], affect collagen synthesis in cultured cells [31,32] and inhibit NADPH oxidase activity [33].…”
