Genetic diversity analysis of Varronia curassavica Jacq. accessions using ISSR markers

ABSTRACT. Varronia curassavica Jacq. is a medicinal and aromatic plant from Brazil with significant economic importance. Studies on genetic diversity in active germplasm banks (AGB) are essential for conservation and breeding programs. The aim of this study was to analyze the genetic diversity of V. curassavica accessions of the AGB of Medicinal and Aromatic Plants of the Federal University of Sergipe (UFS), using inter-simple sequence repeat molecular markers. Twentyfour primers were tested, and 14 were polymorphic and informative, resulting in 149 bands with 97.98% polymorphism. The UPGMA dendrogram divided the accessions into Clusters I and II. Jaccard similarity coefficients for pair-wise comparisons of accessions ranged between 0.24 and 0.78. The pairs of accessions VCUR-001/VCUR-503, VCUR-001/VCUR-504, and VCUR-104/VCUR-501 showed relatively low similarity (0.24), and the pair of accessions VCUR-402/VCUR-403 showed medium similarity (0.78). Twenty-eight accessions were divided into three distinct clusters, according to the STRUCTURE analysis. The genetic diversity of V. curassavica in the AGB of UFS is low to medium, and it requires expansion. Accession VCUR-802 is the most suitable for selection in breeding program of this species, since it clearly represents all of the diversity present in the AGB.

RESUMO:O conhecimento do sistema reprodutivo é fundamental para a conservação e manejo de uma espécie. O objetivo deste trabalho foi descrever a fenologia da floração, a antese, registrar os insetos visitantes no período de floração, determinar as características morfométricas das flores e o sistema reprodutivo da erva-baleeira, em um ambiente de Cerrado do Norte de Minas Gerais. Entre maio a dezembro de 2012 foi caracterizado o comportamento fenológico da floração. Na análise da fenologia floral foi determinado: o crescimento da inflorescência, o número de flores e frutos por inflorescências. Utilizou-se seis acessos que tiveram dez inflorescências marcadas em cada acesso, totalizando 60 inflorescências. A antese foi determinada utilizando quatro inflorescências em duas plantas. Os visitantes florais foram observados in loco e capturados em três dias consecutivos de coleta. As características morfométricas foram determinadas com paquímetro utilizando 20 flores, sendo cinco flores de quatro acessos. Para determinar o sistema reprodutivo utilizou-se a razão pólen:óvulo (P:O), utilizando 50 flores, sendo 10 flores de cinco acessos em pré-antese. Nas condições de Montes Claros, o crescimento das inflorescências de erva-baleeira ocorreu entre meados de agosto e início de outubro, totalizando 45 dias. O florescimento foi observado entre meados de setembro e final de outubro, enquanto a frutificação ocorreu de meados de outubro a início de dezembro, sendo que ambos ocorreram de forma irregular. A antese floral de erva-baleeira, neste estudo, ocorre entre 7:00 e 11:00 horas. Os insetos visitantes pertencem as ordens Coleoptera, Hemiptera, Diptera e Hymenoptera. As flores apresentaram o diâmetro de 2,13 ± 0,05 (mm), o comprimento de 3,29 ± 0,08 (mm), diâmetro do ovário de 0,70 ± 0,02 (mm), o comprimento do ovário de 2,48 ± 0,12 (mm), o diâmetro da antera de 0,67 ± 0,01(mm) e o comprimento da antera de 0,93 ±0,02 (mm), quatro óvulos e cinco anteras por flor. A razão P:O foi de 576,542, indicando que a espécie é alógama facultativa. Morpho-agronomic characterization of Varronia curassavica germplasm conservated “Ex situ”

Varronia curassavica Jacq. is a medicinal and aromatic plant native to Brazil. The essential oil of this species is valued by the pharmaceutical industry due to its bioactive substances containing anti-inflammatory properties. This study aimed to morpho-agronomically characterize 27 accessions of the collection of V. curassavica of the Active Germplasm Bank of Medicinal and Aromatic Plants of the Federal University of Sergipe. The experiment consisted of a randomized block design with three replications. The morphological characterization occurred in the field, by evaluating the variables plant height; canopy width; canopy shape; stem diameterstem color; leaf blades width, length, and length/width ratio; leaves, petals, and sepals color. The agronomic characterization was performed using the essential oils extracted from dried leaves by hydrodistillation in a Clevenger apparatus. Leaves dry matter yield per plant and essential oil yield and content were also evaluated. The accessions showed great variation for leaves color, with different shades of green, which allowed separating them into dark green leaf and light green leaf plants. Plant height values ranged from 101.33 cm (VCUR-801) to 345.33 cm (VCUR-701). The clustering analysis of the traits revealed seven distinct groups. The accessions VCUR-103 and VCUR-001 were the most divergent, whereas the accessions VCUR-401 and VCUR-404 were the most similar. The morpho-agronomic characterization of V. curassavica accessions provides a correct description of the species by pointing out perspectives for genetic improvement, besides optimizing the conservation process.
